急性肝损伤肝肾阴虚证模型的建造和评价 被引量:5

摘要 目的:建立急性肝损伤肝肾阴虚证病证结合大鼠模型。方法:采用长期激怒法制备肝肾阴虚证动物模型,在此基础上使用CCl4原液一次性腹腔内注射,复制CCl4急性肝损伤模型,并从一般状态、饮水量、进食量、游泳时间、肛温、唾液酸碱度、体重减轻、肝脏系数、肝功能、肝脏形态、肝组织病理等方面对模型进行评价。结果:该模型具有阴虚证的典型特征形体消瘦,又具备肝的定位症状肝损伤显著、抗疲劳能力降低和肾的定位症状虚损、尿液异常等,因此,该模型符合动物肝肾阴虚证的辨证标准[7],同时,该模型肝脏系数显著增大(P<0.05),肝功能明显异常(P<0.001),肝组织病理显示肝损伤显著,因此,该模型同时符合急性肝损伤的诊断标准。结论:应用长期激怒法复合CCl4腹腔注射法制备的急性肝损伤肝肾阴虚证大鼠模型,是理想的病证结合动物模型,可用于中医病证病理基础及其治疗的研究。 Objective:To construct disease and syndrome integrated rat model of yin-deficiency syndrome of liver and kidney in the acute hepatic injury.Methods: First,to made rat model of yin-deficiency syndrome of liver and kidney by long irritated method,then to made the acute hepatic injury model by intraperitoneal injection with CCl4.To evaluate the model by the aspects of common state,drink quantity,food quantity,swimming time,anus temperature,saliva PH,lose weight,liver quotiety,liver function,liver configuration and liver pathology.Results: The model has the thin body which belongs to characteristics of yin-deficiency syndrome.Its prominence liver injury and reduced resisting fatigue belong to symptomes of liver and its weak and urine abnormity belong to symptomes of kidney.So,the model accords with the differentiation standard of yin-deficiency syndrome of liver and kidney.Its augmented liver quotiety,abnormity liver function,prominence liver injury by pathology accord with the diagnostic standard of acute hepatic injury.Conclusion:The animal model of yin-deficiency syndrome of liver and kidney in the acute hepatic injury made by long irritated and intraperitoneal injection with CCl4 method is the perfect disease and syndrome integrated rat model,this model can be used in the research of pathology basis and therapia of disease and syndrome of TCM.
