摘要
目的:研究STAT3在脑缺血后的信号转导、调控机制以及电针的干预作用。方法:采用热凝闭大鼠大脑中动脉致局灶性脑缺血模型,采用原位杂交的方法检测不同时间段STAT3mRNA表达水平。结果:相较假手术组,造模2h后各指标出现轻微升高,24h后进一步升高,在多个指标上与假手术组相较有显著性差异(P<0.05)。3天后数值下降。电针组的检测结果与模型组的趋势相同,即2h组开始升高,24h组较高,3天组下降。但与模型组相较,各时间段在数值上均高于相应的模型组,具有一定的优势。结论:电针治疗可以提高脑缺血后STAT3mRNA表达水平,激发机体的自我保护机制。同时,还可以维持这样保护机制持续更长的时间。由此可推断:电针对脑功能的改善作用,可能与其上调STAT3mRNA表达,进而减少病灶区神经细胞的凋亡有一定的关系。
Objective: To study STAT3 signal transduction,regulation and control mechanisms after cerebral-ischemia and the role of electro-acupuncture(EA)intervention.Methods: Focal cerebral ischemia (FCI)model was established by middle cerebral artery occlusion(MCAO)with heat—coagulation method.The expression of STAT3mRNA is detected in the different time periods by using in situ hybridization method.Results: Compared to sham-operated group,after 2 hours each indicator of the model one appears slightly increased,and further increased in a number of indicators after 24 hours,there was significant difference compared with the sham-operated group(P〈0.05).the data decline after 3 days.Electro- acupuncture groups of test results have the same trend with model groups.However,compared with model group,the numerical are higher than the corresponding model group on each time,has certain advantages.Conclusion: EA therapy can increase the expression level of STAT3mRNA after cerebral-ischemia,and stimulate the body's self-protection mechanisms.At the same time,EA therapy can also continue to maintain such protection mechanism longer.Thus we can be inferred: the improvement role of brain function by EA therapy may have a certain relationship with increasing of STAT3mRNA-expression,thereby reducing apoptosis of the damaged neuronal.
