马铃薯茎尖脱毒培养基的筛选及影响因素分析 被引量:8

摘要 以国际马铃薯中心提供的编号为B13-6的马铃薯薯块进行脱毒培养,研究了添加不同激素的培养基和茎尖大小对马铃薯茎尖成活及脱毒效果的影响。研究结果表明培养基"MS+GA30.2mg/L+6-BA0.5mg/L+NAA0.05mg/L+泛酸钙0.2mg/L+肌醇100mg/L"为最佳培养基,不产生愈伤组织,直接成苗;带1个叶原基的茎尖有较高的脱毒率,但成活率较低;而带2个叶原基的茎尖脱毒率较低,但成活率较高。对薯块的处理可降低污染率,得到更多的无菌苗提高脱毒苗的成活率;对薯块进行热处理后可提高脱毒效果。 The virus-free cultivation of potato B13-6 supplied by the International Potato Research Center was carried out, the effect of different hormone medium and different sizes of stem apex on survival and detoxification rate was studied. The results showed that culture medium No.6 (MS+GA3 0.2 mg/L+6-BA 0.5 mg/L+NAA 0.05 mg/L+calcium pantothenate 0.2 mg/L+ inose 100 mg/L) is the best, can grow young plant without callus; the stem apex with one leaf primordium has high detoxification rate, but low survival rate; the stem apex with two leaf primordiums has low detoxification rate, but high survival rate. Process of potato can reduce pollution rate, and also can get more aseptic seedlings. Heattreatment on potato can improve detoxification effects.
