溶血磷脂酸通过LPA1/3受体-ERK-c-myc途径促进间充质干细胞增殖 被引量:1

摘要 目的移植至梗塞心脏的间充质干细胞存活数量少限制了其治疗效果。本研究旨在检测溶血磷脂酸对间充质干细胞促增殖作用,以及这种作用的机制。方法大鼠间充质干细胞培养、传代及处理,Brdu合成掺入法检测细胞增殖,加入特异性抑制剂验证相关信号通路,细胞蛋白提取及western blot检测蛋白表达。结果溶血磷脂酸促进间充质干细胞增殖,并呈浓度依赖性。LPA1/3受体特异性抑制剂,ERK信号通路抑制剂都不同程度降低了溶血磷脂酸的促增殖作用。溶血磷脂酸的作用使细胞内c-myc蛋白表达上调,而加入上述抑制剂则下调了c-myc蛋白的表达。结论溶血磷脂酸能够促进间充质干细胞增殖,这种作用是通过结合LPA1/3受体,活化ERK信号通路,从而上调c-myc蛋白表达实现的。 Objective To investigate the effect of Lysophosphatidic acid on mesenchymal stem cells proliferation in order to improve the amount of the cells transplanted in infracted heart. Methods Mesenehymal stem cells were obtained, cultured and treated. The proliferation of the cells was measured by BrdU incorporation during DNA synthesis. The inhibitors were used to testify relative signaling pathways, and proteins were obtained and detected by western blot. Results Lysophosphatidic acid promoted proliferation of mesenehymal stem cells in a dose-dependent manner. The inhibitors for LPA1/3 receptor and ERK prevented the cell proliferation. Lysophosphatidic acid also increased the expression of c-mye, which was inhibited by the two inhibitors. Conclusion Lysophosphatidic acid promotes proliferation of mesenchymal stem cells by binding LPA1/3 receptor, activating ERK pathway and increasing the expression of c-mye.
