柑橘黄酮对内毒素诱发大鼠乳腺炎的抑制作用 被引量:3

摘要 SD大鼠产后经乳头管注入大肠杆菌内毒素诱发大鼠乳腺炎症,观察不同剂量的柑橘黄酮(200,600mg.kg-1.d-1)的抗乳腺炎作用.乳腺组织病理学观察表明:高剂量(600 mg.kg-1.d-1)的柑橘黄酮预处理能有效地抑制内毒素诱发的大鼠乳腺炎症反应,明显减少乳腺组织的炎性细胞浸润,使乳腺组织结构清楚;并能抑制内毒素诱导的乳腺炎母鼠血清碱性磷酸酶水平升高(P<0.01).体外抑菌实验显示:柑橘黄酮对金黄色葡萄球菌有很强的抑制作用,对大肠埃希菌和铜绿假单胞菌也有较强的抑制作用.以上结果提示柑橘黄酮可抑制乳腺炎的发生,该作用可能与其抑制炎性反应和抑菌功能有关. The inhibitive effects of citrus flavonoids on mastitis induced by endotoxin via teat duct in rat was studied.Female SD rats were administrated citrus flavonoids(200 or 600 mg·kg+-1·d+-1) by ig from gestation day 1 to day 4 after parturition.Escherichia coli endotoxin was inoculated into the fourth(abdominal) mammary gland via the teat duct 72 h after parturition.Exactly 24 h after inoculation,the rats were sacrificed,and the mammary glands were removed and fixed for histopathologic evaluations.Meanwhile,the activity of alkaline phosphatase(AKP) in the serum was detected.Furthermore,the minimal inhibitory concentrations(MIC) of cirus flavonoids to 3 strains of common bacteria was measured.Histopathologic examination of mammary gland revealed that high-dose(600 mg·kg+-1·d+-1) of cirus flavonoids can inhibit inflammatory reaction induced by Escherichia coli endotoxin.The number of polymorphonuclear neutrophi infiltrated in alveoli was decreased.AKP activity of serum in mastitis model rats was significantly reduced by high-dose(600 mg·kg+-1·d+-1) of cirus flavonoids(P0.01).Bacteriostatic test in vitro showed that cirus flavonoids had inhibitive effects on Staphylococcus auerus,Escherichia coli,and Pseudomonas aeruginosa.The results suggested that citrus flavonoids had inhibitive effects on mastitis,anti-inflammatory and bacteriostatic action might be involved.
