组织因子对奥沙利铂抑制人胃癌细胞侵袭及诱导其凋亡的影响

Effect of tissue factor on invasion inhibition and apoptosis inducing effect of oxaliplatin in human gastric cancer cell

目的观察组织因子(TF)对奥沙利铂抑制人胃癌细胞侵袭及诱导其凋亡的影响。方法采用已成功转染TF-pcDNA3重组体(实验组)及空载体pcDNA3(对照组)的人胃癌细胞株SGC7901,与奥沙利铂共同孵育。采用Transwell小室观察细胞侵袭能力,采用比色法检测Caspase-3活性,采用Annexin-V/PI双染色法检测细胞凋亡率。结果与对照组比较,实验组细胞侵袭能力增高,Caspase-3活性及细胞凋亡率降低,P均<0.05。结论TF表达增高可降低奥沙利铂对胃癌细胞体外侵袭力的抑制作用,减弱奥沙利铂诱导的细胞凋亡。

Objective To observe the effect of tissue factor（TF）on invasion inhibition and apoptosis inducing effect of oxaliplatin in human gastric cancer cell line SGC7901.Methods SGC7901 cells wich has been successfully transfected with TF-pcDNA3 recombinant（Experimental group）and empty vector pcDNA3（Control group） were incubated with oxaliplatin.Transwell chamber were used to show the change of invasive ability of cells.Caspase-3 activity were detected by colorimetric method;Annexin-V/PI double-staining method was used to detect the apoptosis rate.Results Compared with the control group,invasion ability increased but Caspase-3 activity and apoptosis rate decreased in the experimental group,P0.05.Conclusion TF can reduce the invasion inhibition and apoptosis inducing effect of oxaliplatin in human gastric cancer cell in vitro.
