木薯新品种新选048离体微扦插快速繁殖技术 被引量:4

摘要 以木薯新品种新选048腋芽为外植体,对木薯离体微扦插途径进行研究。结果表明,采用0.1%HgCl2溶液灭菌木薯外植体6~12 min,对外植体的存活率影响不明显,外植体成活率为32.3%~37.9%。将在MS+NAA 0.1 mg/L中诱导培养获得的腋芽剪成带芽茎段,扦插到含0~0.3 mg/L NAA、0~1.5 mg/L 6-BA的MS培养基中,其在MS+0.3 mg/L NAA培养基中的增殖和生长效果最好,增殖系数高、苗壮且整齐、根白色粗壮,愈伤组织白色、生长正常。与河沙相比,以木糠为木薯组培苗的假植基质较好,成活率达70%以上。 The axillary buds of new cassava variety Xinxuan 048 were used as explants to study the miero-euttage in vitro culture and rapid propagation technology for cassava. The results showed that disinfection with 0.1% solution of mercuric chloride on the explants of cassava for 6-10 min showed no significant effect on survival rate of explants which ranged from 32.3% to 37.9%. The shoots with one axillary bud induced from MS+NAA (0.1 rag/L) medium were cuttaged to MS medium with different hormone combinations ranging from 0-0.3 mg/L NAA and 0-1.5 mg/L 6-BA. It was found that MS+NAA 0.3 mg/L was the best medium for the propagation and growth of shoots and resulted in high multiplication coefficient, vigorous plantlets and white roots and normal white callus. For temporary planting of the plantlets, the wood chaff was found to be the best with more than 70% of survival rate.
