摘要
目的检测1,25二羟维生素D3〔1,25(OH)2D3〕对人子宫内膜癌HEC-1-A细胞作用后S期激酶相关蛋白(Skp2)表达情况,以探讨其抗癌机制;方法选用人子宫内膜癌细胞株HEC-1-A进行体外培养,采用RT-PCR在mRNA水平检测Skp2基因表达的变化;结果 (10-8-10-6)mol.L-1浓度的1,25(OH)2D3作用于HEC-1-A细胞随着作用浓度增加、作用时间延长,细胞增殖能力逐渐下降,Skp2mRNA逐渐减少;结论 1,25(OH)2D3对人子宫内膜癌细胞株HEC-1-A有抑制作用,可能通过转录水平下调Skp2基因表达,从而产生抑癌作用。
OBJECTIVE To study the mechanism of 1, 25 (OH)2D3 on human endometrial carcinoma cell HEC-1-A by detecting the expression of S-phase kinase associated protein 2 (Skp2) gene. METHODS Human endometrial carcinoma cells HEC-1-A were cultured in vitro and the expression level of Skp2 mRNA was examined by reversed transcript polymerase chain reaction (RT-PCR). RESULTS The results showed that (10^-8-10^-6)mol·L^-1 1,25(OH)2D3 inhibited the proliferation of HEC-1-A cell line, and the mRNA expression of Skp2 in HEC-1-A cell lines after 1,25 (OH)2D3 treatment decreased with concentration of 1,25(OH)2D3 and treatment time. CONCLUSION 1,25(OH)2D3 can inhibit the growth of HEC-1-A cell line. It may inhibit cancer cell proliferation through down-regulation of Skp2 gene.
