腺病毒介导的nm23-H1对人结直肠癌裸鼠移植作用的实验研究

摘要 目的探讨Ad-GFP-nm23-Hl对人高转移结直肠癌移植瘤的抑制作用,为临床肿瘤基因治疗提供一定的理论依据。方法将15只人结直肠癌移植瘤裸鼠随机分为高剂量组(1010PFU/mlAd-GFP-nm23-Hl)、低剂量组(109PFU/mlAd-GFP-nm23-Hl)、阴性对照组(PBS)。均采用瘤体注射给药,连续给药4d,每2d测一次肿瘤体积。观察10d后取瘤块称重和组织病理学检查。结果Ad-GFP-nm23-Hl高剂量组移植瘤体积较其他各组明显缩小(P<0.05),瘤体积抑制率和瘤重抑制率分别为76%和66%。结论Ad-GFP-nm23-Hl对人高转移结直肠癌移植瘤有抑制作用,并存在剂量依赖性。 Objective To study the inhibitory effects of Ad-GFP-nm23-Hl on human colorectal carcinoma bearing nude mice and to gain further insight into some theoretical and methodical basis for using nm23-Hl gene therapy in cancers. Methods Fifteen nude mice inoculated subcutaneously with human colon cancer cell line Lovo,were allocated randomly into 3 groups:high dose (treated with 1010PFU/ml Ad-GFP-nm23-Hl),low dose (treated with 109 PFU/ml Ad-GFP-nm23-Hl) and control group (treated with PBS). Mice were injected uninterruptedly for 4 days. Tumor volume was measured every three days. The animals were killed on the sixth day after treatment,the xenografts were weighed,and studied histologically. Results The volume and weight of xenografted cancer in Ad-GFP-nm23-Hl groups were markedly lower than those in control group. Tumor volume and weight inhibition rates of the Lovo cell line which were treated with Ad-GFP-nm23-Hl of 1010PFU/ml were 76% and 66% respectively. Conclusions Ad-GFP-nm23-Hl showed significant inhibitory effects on proliferation of human colorectal carcinoma Lovo cell line in vivo.
