摘要
构建了圆斑星鲽肌肉和脾脏组织的cDNA文库,2个文库的库容量分别为5.0×106和1.1×106,重组率为97.3%和93.2%,平均插入片断长度大于800 bp。进行了1002个(肌肉541个,脾脏461个)ESTs测序,得到621个(肌肉192个,脾脏429个)单基因簇,其中包括110个(肌肉87个,脾脏23个)重叠群和511个(肌肉105个,脾脏406个)单拷贝。BLASTX分析的结果显示,318(51.2%)个基因簇有相关同源性,其他的303个EST(48.8%)无明显的同源性(E值≤E-5)。
cDNA libraries of muscles and spleen in healt constructed. The titer of the libraries was 5.0× 10^6 for combined efficiency of each eDNA library exceeded 97.3 spotted halibut＇s（Verasper variegatus） were muscles and 1.1×10^6 for the spleen. The reand 93.2%. The average insert size was over 800 kb. Total of 1002（muscle.578, spleen.480）ESTs were sequenced and analyzed, and 621 unigenes involving 110 contigs and 511 singletons were annotated by BLASTX. Of the unigenes, 318（51.2%）were assigned putative functions（E≤1.00E^-5）.
