酿酒酵母CHS3基因缺失对热耐受性的影响 被引量:5

摘要 研究真菌细胞壁几丁质合成酶CHS3基因在酵母热耐受性中的功能。采用长臂同源多聚酶链式反应(LFH-PCR)技术构建酵母CHS3基因缺失菌株;比较野生型与CHS3基因缺失菌株的几丁质含量;梯度生长实验检测CHS3基因缺失菌株的热敏感性。结果显示,与野生型菌株相比,CHS3基因缺失菌株表现出热胁迫敏感性表型,此表型可被抗氧化剂N-乙酰半胱氨酸(NAC)弥补,表明该热敏感表型可能与热胁迫条件下活性氧积累引发的氧化损伤。 This paper was to investigate the role of chitin synthase gene CHS3 in yeast heat resistance. The CHS3 gene deletion mutant was construct- ed by long flanking homology PCR (LFH-PCR). The chitin content of wild type and transformants were compared. The heat sensitivity of CHS3 gene deletion mutant was investigated by spot dilution growth assays. The results showed that the CHS3 gene deletion mutant was more sensitive to the heat stress than the wild type, which could be restored by the addition of the antioxidant NAC. The increased heat sensitivity of CHS3 gene deletion mutant may be caused by the accumulation of reactive oxygen species under heat stress condition, which can lead to oxidative damage.
