摘要
建立测定玉米浆中游离氨基酸的反相高效液相色谱法。DiamonsilCl8(2)反相柱(5μm,250mm×4.6mm):梯度洗脱,流动相A相:0.05mol/L醋酸钠缓冲溶液(pH6.50),B相:乙腈-水(v/v=1:1),流速1.0mL/min,检测波长360nm。在该条件下所得各氨基酸的线性回归方程相关系数均在0.999以上。测定17种氨基酸的加样回收率94%-105%,相对标准偏差小于2.0%。
Method of determination of flee amino acids in corn syrup with RP-HPLC was established. The detection conditions were: using re- versed-phase column Diamonsil C18 (2) (5 μn, 250 min×4.6 mm); gradient elution and mobile phase A: 0.05 mol/L sodium acetate buffer solution (pH = 6.50); phase B: methyl cyanide : water by 1:1 (V/V), flow rate: 1.0 ml/min, detection wavelength: 360 nm. Under the conditions, correlation coefficients of linear regression equations of amino acids could reach above 0.999. Recovery of 17 kinds of amino acids determination was in the be- tween 94-105% and relative standard deviation was less than 2.0%. The method is simple and reliable. It can be used in determination office amino acids in corn syrup for antibiotic fermentation.
