摘要
目的:观察脑缺血大鼠大脑皮质胰岛素样生长因子II(IGF-II)免疫阳性神经元的变化,并探讨复方丹参的抗缺血机制。方法:双侧颈总动脉结扎建立永久性脑缺血模型,复方丹参腹腔内注射后用ABC法显示IGF-II免疫阳性神经元并进行定量分析。结果:大鼠大脑皮质II-V层有较丰富的IGF-II免疫阳性神经元分布,阳性细胞多见一个细长的突起。对照组大鼠阳性神经元分布较均匀、不密集,60、120、180 d光密度无明显变化;脑缺血组大鼠60 d时密集,较对照组增多明显,光密度增高,但120、180 d数量有所减少,光密度下降,但仍多于对照组。生理盐水处理组IGF-II免疫阳性神经元的变化与单纯缺血大鼠对应时间点相似,用丹参处理后60 d,阳性神经元的数量较缺血组和生理盐水处理组数量减少,光密度未见明显差异,120 d时数量仍然较密集,多于对照组,180 d进一步减少,接近对照组,但多突起的阳性细胞多见。结论:脑缺血性刺激可诱导大鼠大脑皮质IGF-II免疫阳性神经元增多,丹参干预后IGF-II免疫阳性神经元未见明显增多,但表达IGF-II免疫阳性神经元形态特征发生变化。
Aim:To observe the changes of insulin-like growth factors-Ⅱ immunopositive neurons in the cerebral cortex of cerebral ischemia rats and the anti-ischemia mechanism of Danshen. Methods: Firstly cerebral ischemia rats were established by permanent bilateral carotid artery ligation,then Danshen was injected into peritoneal cavity.IGF-Ⅱ immunopositive cells were stained by ABC method,and the number of them was quantitatively analyzed by image analyzer.Results: Many IGF-Ⅱ immunopositive neurons distributed in Ⅱ-Ⅴ layer of cerebral cortex and lmost of them had only one neurite being stained.In the control group,the immunopositive neurons were not intensive in number and no significant change in optic density was found in every phase.However,after 60 days the artery ligation in cerebral ischemia rats more neurons were found in the cerebral cortex and optic density was higher than that in 120 days and 180 days.The optic density in 120 days and 180 days was decreased slowly,but was still higher than that of control group.The changes of immunopositive cells in rats treated with NS are similar to cerebral ischemia rats.After rats were treated with Danshen,the immunopositive cells in 60 days did not increase like that in ischemia group showing more immunopo sitive neurons in the cerebral cortex,and no significant changes in density was found.The number of immunopositive cells in 120 days was decreased compared with that in 60 days,but optic density was higher than that in control group.In 180 days,optic density was down to the level of the control group.Meanwhile,the cellular shape was changing and the pin-like cells were diminished,and immunopositive cells with more neurites were stained.Conclusion:Stimulation of cerebral ischemia could induce the increase of IGF-Ⅱ immunopositive neurons in number.After treated with Danshen these immunopositive cells could not grow in number,however,their characters were changed.
