真菌蛋白酶对人支气管上皮细胞系BEP2D分泌细胞因子的影响

摘要 目的探讨真菌蛋白酶对人支气管上皮细胞系BEP2D细胞分泌细胞因子的影响。方法分别用0、1、2和4μmol/mL的真菌蛋白酶刺激人支气管上皮细胞系BEP2D细胞,共同培养6和12 h后收集细胞,用免疫印迹(western blotting)方法检测内皮细胞表达人胸腺基质淋巴细胞生成素(TSLP)蛋白水平,用定量PCR方法检测胸腺活化调节因子(TARC)、白介素(IL)-12和α肿瘤坏死因子(TNF-α)的mRNA表达。结果人支气管上皮细胞系BEP2D细胞TSLP蛋白及TARCmRNA表达增加,并呈剂量依赖关系,但IL-12和TNF-α表达水平无显著改变。结论真菌蛋白酶能刺激支气管上皮细胞BEP2D分泌与哮喘相关的细胞因子,其可能是真菌诱发哮喘的机制之一。 Objective To observe the effect of fungus protease on the cytokines production of human bronchus epithelium cell line BEP2D. Methods Fungus proteases (0, 1, 2 and 4 μmol/mL) stimulated human bronchus epithelium cell line BEP2D and cultured cells were collected after 6 and 12 h. The level of TSLP protein was detected by western blotting, mRNA of cytokines (TARC, IL-12 and TNF-α) was observed by real-time PCR. Results The level of TSLP protein and mRNA of TARC was increased dose-dependently. But there was no significant change in the level of IL-12 or TNF-α. Conclusion Fungus protease can induce the expression of cytokines associated with asthma in bronchus epithelium cell line BEP2D, which may be one of the asthma pathogenesis by fungus protease.
