三参保肺颗粒对慢阻肺合并肺纤维化大鼠肺组织Ⅰ、Ⅲ型胶原及转化生长因子β_1表达的影响 被引量:4

摘要 目的探讨三参保肺饮颗粒对慢阻肺合并肺纤维化大鼠肺组织Ⅰ、Ⅲ型胶原及转化生长因子β1(TGF-β1)表达的影响。方法将40只w istar大鼠随机分为对照组、模型组、强地松组、三参组,用熏香烟烟雾加气管内注入弹性蛋白酶和博来霉素方法建立大鼠COPD合并肺纤维化模型。对照组造模时气管内注入生理盐水,强地松组、三参组于造模完成后予强地松、三参保肺饮灌胃,用免疫组织化学方法观察各组病理切片支气管肺组织TGF-β1蛋白表达,用天狼猩红染色观察各组支气管壁Ⅰ、Ⅲ型胶原分布,用病理图像分析系统测量各组切片支气管肺组织TGF-β1免疫组化阳性细胞平均面积、积分光密度和平均光密度,比较各组支气管壁Ⅰ、Ⅲ型胶原面积、面密度、光密度。结果三参组阳性细胞平均面积较其他三组显著增加(P<0.01);积分光密度、平均光密度较模型组显著降低(P<0.01);平均光密度较强地松组降低(P<0.05)。模型组、三参组Ⅰ、Ⅲ型胶原阳性目标面积较对照组显著增加(P<0.05;P<0.01);三参组较模型组显著降低(P<0.05);三参组Ⅰ、Ⅲ型胶原面密度、光密度较模型组显著降低(P<0.05),但两组较对照组显著增加(P<0.05,P<0.01)。结论三参保肺颗粒有下调慢阻肺合并肺纤维化大鼠肺组织Ⅰ、Ⅲ型胶原及TGF-β1表达,减轻肺纤维化的作用。 Objective To investigate the impact of sanshen baofei granules on collagen Ⅰ and Ⅲ as well as TGF-β1 expression in lung tissue of rats with chronic obstructive pulmonary disease(COPD).Methods 40 Wistar rats were randomly divided into control group,model group,prednisone group and sanshen group.The model of COPD and pulmonary fibrosis was established in mice by cigarette smoking and intratracheal infusion of elastase and bleomycin.In control group,physiological saline was infused intratracheally,in prednisone group and sanshen group,after modeling,gastric infusion of prednisone and sanshan baofei granules were given respectively.IHC method was used to observe TGF-β1 protein expression in bronchial pulmonary tissue of pathologic slices in each group.Sirius rosa BB staining was applied to observe the distribution of collagen Ⅰ and Ⅱ on bronchial wall.Pathological imaging analysis system was used to measure mean area of TGF-β1 IHC positive cell,integral optical density and mean optical density of bronchial pulmonary tissue of pathologic slices in each group.The area of collage Ⅰ and Ⅱ,surface density and optic density of bronchial wall were compared among groups.Results The mean area of positive cell increased significantly in sanshen group as compared with other three groups(P〈0.01);the integral optical density and mean optical density decreased significantly as compared with model group(P〈0.01),the mean optical density decreased as compared with prednisone group(P〈0.05).The positive target area of collage Ⅰ and Ⅱ in model group and sanshen group increased significantly as compared with control group(P〈0.05,P〈0.01)and that in sanshen group decreased significantly as compared with model group(P〈0.05).The surface density and optical density of collagen Ⅰ and Ⅱ in sanshen group decreased significantly as compared with model group(P〈0.05).But those in two groups increased significantly as compared with control group(P〈0.05,P〈0.01).Conclusion Sanshen baofei granules down-regulate collagen Ⅰ and Ⅱ as well as TGF-β1 expression in pulmonary tissue of mice with COPD and pulmonary fibrosis and alleviate pulmonary fibrosis.
