mPEG5000修饰的VLA-4拮抗肽KILDVA对致敏性哮喘小鼠肺组织Eotaxin表达的影响

摘要 观察mPEG5000修饰的VLA-4拮抗肽KILDVA对小鼠哮喘性气道炎和肺组织CC型趋化因子Eotaxin表达的影响,探讨mPEG5000-KILDVA抗支气管哮喘的作用机制.采用卵清白蛋白(OVA)建立小鼠哮喘模型.观察小鼠的行为变化,制备小鼠血清,收集肺泡灌洗液进行白细胞总数和嗜酸性粒细胞计数;采用酶联免疫吸附法测定血清、支气管肺泡灌洗液中Eotaxin水平;免疫组化方法检测肺组织Eotaxin的表达.结果显示:模型组小鼠血清和支气管肺泡灌洗液中Eotaxin水平较正常组明显升高(p<0.01);mPEG5000-KILDVA组比模型组明显减少(p<0.05);mPEG5000-KILDVA组支气管与地塞米松组无显著差异(p>0.05).模型组小鼠支气管肺泡灌洗液中白细胞总数及嗜酸性粒细胞计数较正常组明显增加(p<0.01);mPEG5000-KILDVA组较模型组明显下降(p<0.05);mPEG5000-KILDVA组支气管与地塞米松组无显著差异(p>0.05).模型组肺组织Eotaxin蛋白表达量较正常组明显升高(p<0.01),mPEG5000-KILDVA组小鼠较模型组明显下降(p<0.05);mPEG5000-KILDVA组支气管与地塞米松组无显著差异(p>0.05).研究提示mPEG5000-KILDVA减轻哮喘小鼠的气道炎症与抑制哮喘小鼠肺组织Eotaxin的过度表达有关. To observe the effects of mPEG5000-KILDVA on the antigen-induced airway inflammation and the expression of CC cheomokine Eotaxin in mice asthma models.Approach the mechanisms of mPEG5000-KILDVA on anti-asthma.The mice asthma models were established with ovalbumin(OVA),to observe the change of behavior in mice,prepared serum of mice and collected cbronchoalveolar lavage(BAL) to perform counts of Leukocyte and EOS.The levels of Eotaxin in serum and BALF were measured by ELISA.The expression of Eotaxin in lung tissue were detected by immunohistochemistry.The Eotatin levels of serum and BALF in model group were significantly higher than that in normal control group with significant difference(p〈0.01),mPEG5000-KILDVA group is significantly lower than that in the model group with significant difference(p〈0.05),mPEG5000-KILDVA group and dexamethasone group are similar with no significant difference(p〉0.05).The Eotaxin protein expression unit(PU) is significantly higher in the model group than that in the normal control group with significant difference(p〈0.05),mPEG5000-KILDVA group and dexamethasone group are similar with no significan (p〉0.01).Total cell counts,the absolute number of Leukocyte and EOS in BALF in model group significantly increased compared with normal control group with significantly difference(p〈0.01),mPEG5000-KILDVA group is significantly lower than that in the model group with significant difference(p〈0.05),mPEG5000-KILDVA group and dexamethasone group are similar with no significant difference(p〉0.05).mPEG5000-KILDVA can reduce airway inflammation and inhibit Eotaxin excessive expression of asthmatic mouse.
