大鼠坐骨神经切断后相应脊髓节段Bcl-2、Bax蛋白表达及神经细胞凋亡 被引量:2

Changes of Cell Apoptosis and the Expressions of Bcl-2 and Bax in Spinal Cord after the Transection of Rat Sciatic Nerve

摘要 目的探讨成年大鼠坐骨神经切断后相应脊髓节段Bcl-2、Bax蛋白表达及神经细胞凋亡。方法选取健康成年雄性Wistar大鼠35只,随机分为假手术组(5只)和实验组(30只)。实验组再分为六个小组,每组5只。实验组于伤后3天,1周,2周,4周,6周和8周6个时间点取材。应用TUNEL法和免疫组化染色法检测神经细胞的凋亡及脊髓Bcl-2、Bax蛋白表达。结果大鼠坐骨神经切断后3天即可见脊髓内TUNEL阳性细胞,2周表达到达高峰,此后表达量逐渐下降;大鼠坐骨神经切断3天后凋亡相关蛋白Bcl-2和Bax出现表达,2周时达高峰,8周时仍有表达。Bcl-2/Bax值2周时最低,以后逐渐增大,Bcl-2/Bax值越小凋亡的细胞越多。结论大鼠坐骨神经切断后相应脊髓节段内存在神经细胞的凋亡,Bcl-2和Bax比值的变化是神经凋亡的重要指标。 Objective To study the changes of cell apoptosis and the expressions of bcl-2and Bax in spinal cord after the transection of adult rat sciatic nerve.Methods Thirty-five Wistar male rats were randomly divided into sham operated group(5) and experimental group(30).The experimental group was subdivided randomly into 6 groups,5 in each group.The spinal cord in the experimental group was taken at 3 d,1 w,2 w,4 w,6 w and 8 w after injury.The terminal deoxynucleotidal transferase-mediated dUTP-biotin nick end labeling(TUNEL) method was used to detect the neural cell apoptosis and immunohistochemical method was used to observe the expressions of Bcl-2 and Bax.Results TUNEL-positive cells were seen prmiarily at 3d after injury,peaked at 2w and then decreased,but few positive cells still exited at 8w.The Bcl2/Bax ratio kept minmial at 2w and then gradually increased.The lower the Bcl-2/Bax ratio was,the more there were apoptotic cells.Conclusion The apoptosis of neural cells and the expressions of Bcl-2 and Bax occur after the transection of sciatic nerve.The Bcl-2/Bax ratio is related to the neural cell apoptosis after the transection of sciatic nerve in rats as an important indicator.
