摘要
目的探讨12-脂氧化酶(12-LO)对大鼠肾小球内血管紧张素转换酶(Angiotensin-converting enzyme,ACE)和血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)表达水平的影响。方法用12-LO作用产物12(S)-HETE刺激正常大鼠,观察大鼠肾小球内ACE和AngⅡ的变化。采用ELISA、Western blot和RT-PCR分别检测目标基因mRNA和蛋白的表达。结果 12(S)-HETE使大鼠血糖、24小时尿蛋白略升高,增加肾小球内ACE mRNA和AngⅡ蛋白的表达(P<0.05),但对大鼠体重、肾重/体重无明显影响。结论 12-LO上调大鼠肾小球内ACE和AngⅡ的表达水平。
Objective To investigate the effect of 12-lipoxygenase（12-LO） on the expressions of ACE and AngⅡin normal rats glomeruli.Methods ACE mRNA and AngⅡ protein expression were determined by the product of 12-lo（12（S）-HETE） to stimulate the normal rats.Enzyme-linked immuno sorbent assay,Western blot and reverse transcription-polymerase chain reactions were used for evaluating mRNA and protein expression respectively.Results ACE and AngⅡlevels were increased after 12（S）-HETE stimulation compared with control（P0.05）.However,urine protein,serum glucose,Kidney/weight index were not affected by 12（S）-HETE stimulation.Conclusion 12-LO can upregulate the levels of ACE and AngⅡ in glomeruli.
