摘要 目的 建立血浆样品中万古霉素测定的高效液相色谱法,为临床监测血药浓度以调整万古霉素剂量提供方法 学基础.方法 血浆样品通过10%硫酸锌沉淀,采用C18(250 min×4.6 mm,μm)色谱分析柱,以乙腈-0.05 mol·L^-1磷酸二氢钾缓冲液(pH3.20,10:90)为流动相,流速为1.0 mL·min^-1,检测波长为236 nm,柱温为25℃.进样量为10μL.结果万古霉素血浆样品质量浓度在2.08~83.0 mg·L^-1范围内呈良好的线性关系(r=0.999 5,n=7).高、中、低3个质量浓度溶液的提取回收率分别为97.3%,92.1%和92.8%,RSD分别为2.2%,4.5%和9.0%;其方法 回收率分别为101.2%,95.6%和99.2%,RSD分别为6.0%,4.5%和8.5%.结论 方法 灵敏、快速、简便、可靠,可用于万古霉素的血药浓度监测及临床药学研究. Objective To establish a high performance liquid chromatography method to determine the concentration of vancomycin in human plasma. Methods The vancomycin,extracted from the plasma by sedimentation with 10% zinc sulfate solution,was determined by C18 ( 250 mm×4.6 mm, 5 μm) column with a mixture of acetonitrile and 0.05 mol·L^-1 potassium dihydrogen phosphate (10 : 90} as mobile phase, The elution rate was 1.0 ·mL min^-1 , the injection volume was 10 μL,the detection wavelength was 236 nm,and the column temperature was 25 ℃. Results The linear range for vancomycin was from 2.08 to 83.0 mg· L^-1 with a good correlation coefficient of 0. 999 5( n = 7 ). The extraction recoveries of high, middle and low concentrations were 97. 3 %, 92.1% and 92.'8% with RSD 2.2% ,4.5% and 9.0% ,respectively. The method recoveries were 101.2%, 95.6% and 99.2% with RSD 6.0%, 4.5% and 8.5% ,respectively. Conclusion The method is sensitive, rapid,simple and accurate. It could be applied to monitor the concentration of vancomycin in plasma,and can be used to the clinical pharmaceutical research.
