摘要
目的:观察沙苑子黄酮对人肝癌HepG2细胞的体外抑制作用。方法:采用MTT法观察沙苑子黄酮对人肝癌HepG2细胞的抑制作用,进一步观察沙苑子黄酮对HepG2细胞形态、超微结构、生长曲线以及集落形成的影响。结果:沙苑子黄酮可明显抑制HepG2肿瘤细胞的增殖和集落的形成,50μg/mL剂量组集落形成率为25.6(P<0.01),远低于对照组;光镜及透射电镜可以发现细胞形态有凋亡特征变化。结论:沙苑子黄酮体外对人肝癌HepG2细胞具有明显的抑制肿瘤细胞增殖的作用,其作用与诱导肿瘤细胞凋亡有关。
Objective:To observe the inhibiting effect of flavonoids of Astragalus complanatus(FAC)on human hepatoma carcinoma cell HepG2.Methods:The inhibiting effect on human hepatoma carcinoma cell HepG2 was observed by MTT,growth curve assay and colony formation assay.Its effect on morphology and ultramicrostructure of HepG2 was observed by light microscope and transmission electron microscope.Results:The colony formation ratio is low compared with control group(P0.01),colony formation ratio of FAC-d (50μg/mL) is 25.6%;apoptotic characteristic could be find by light microscope and transmission electron microscope.Conclusion:FAC could markedly inhibit the growth of HepG2 tumor cells in vitro.Its effect may be related to inducing the apoptosis.
