摘要
【目的】观察穴位经皮给药药贴对实验性哮喘豚鼠血清中环氧化酶2(COX-2)水平的影响。【方法】将48只豚鼠随机分为正常对照组、模型组、地塞米松组和经皮给药药贴组,每组各12只。除正常对照组外,各组豚鼠均采用卵白蛋白致敏法复制实验性过敏性哮喘模型。经皮给药药贴组采用大椎、肺俞、肾俞穴位贴敷治疗,地塞米松组采用腹腔注射地塞米松0.5 mg/kg治疗。采用酶联免疫吸附法检测各组豚鼠血清COX-2水平,采用苏木素—伊红染色检测各组豚鼠支气管肺组织病理形态变化。【结果】模型组豚鼠血清COX-2水平显著高于正常对照组(P<0.05),经皮给药药贴组、地塞米松组血清COX-2水平显著低于模型组(P<0.05)。模型组豚鼠支气管肺组织出现明显病理损害,经皮给药药贴组支气管肺组织仅出现轻度病理改变。【结论】穴位贴敷经皮给药药贴可能通过降低血清中COX-2水平,从而对哮喘产生治疗作用。
∶Objective To observe the effect of acupoint application of transdermal patch on the serum level of cyclooxygenase-2(COX-2)in guinea pigs with allergic asthma.Methods Forty-eight guinea pigs were equally randomized into four groups: normal control group,model group,dexamethasone group and acupoint application group.Experimental allergic asthma was induced by the method of ovalbumin challenge in the guinea pigs except(Continued on page 241) for the normal control group.Acupoint application of transdermal patch on Dazhui,Feishu and Shenshu was given to acupoint application group,while dexamethasone treatment group was injected with dexamethasone(0.5 mg/kg) intraperitoneally.Serum COX-2 level in the guinea pigs was detected with enzyme-linked immunosorbent assay,and the pathological changes of the bronchus and lung were examined with hematoxylin-eosin staining.Results The serum level of COX-2 in the model group was significantly higher than that in the normal control group(P0.05).After treatment,serum COX-2 level in acupoint application group and dexamethasone group was significantly lower than that in the model group(P0.05).Obvious pathological changes were found in the bronchus and lung of the model group,while were relieved in acupoint application group.Conclusion Acupoint application exerts certain therapeutic effect on bronchial asthma,and its therapeutic mechanism is probably related with lowering serum COX-2 level.
