疏肝活血法对下肢缺血模型大鼠血管内皮祖细胞的影响

Effect of Therapy of Soothing Liver and Activating Blood on Endothelial Progenitor Cells in Peripheral Blood Vessels of Rats with Lower Limb Ischemia

【目的】观察疏肝活血法对下肢缺血模型大鼠外周血管内皮祖细胞(EPC)的影响。【方法】将50只SD大鼠随机分为3组:中药组20只,模型组20只,假手术组10只。中药组和模型组均采用右下肢股动脉结扎及分支剔除术复制大鼠下肢缺血模型,假手术组仅暴露股动脉但不结扎股动脉及其分支。中药组术后按0.5 g.kg-1.d-1剂量灌服柴胡疏肝散合大黄虫丸煎剂,连续7 d。检测各组术前1 d、术后第1、3、7天4个时间点大鼠外周血酶学指标肌酸激酶(CK)、谷草转氨酶(AST)、乳酸脱氢酶(LDH)、α-羟丁酸脱氢酶(HBDH)水平变化及双荧光阳性细胞[CD34+、血管内皮生长因子受体(VEGFR2+)]比例。【结果】术后模型组外周血酶学指标AST、CK、LDH、HBDH水平较假手术组均显著上升,术后第3、7天中药组各项酶学指标水平较模型组及术后第1天均显著下降(P<0.05)。术后中药组和模型组反映EPC变化的CD34+、VEGFR2+双荧光阳性细胞比例较假手术组均显著升高(P<0.05),且中药组升高较模型组更为显著(P<0.05或P<0.01)。【结论】疏肝活血法可有效地改善下肢缺血状态,其作用可能与其能增加外周血EPC数量有关。

Objective To investigate the effect of therapy of soothing liver and activating blood on endothelial progenitor cells（EPC） in peripheral blood vessels of rats with lower limb ischemia.Methods Fifty SD rats were randomized into 3 groups： herbs group（N=20）,model group（N=20） and pseudo-operation group（N=10）.The rats in herbs group and model group were given ligation of right lower limb femoral artery and extirpation of the artery branch to induce lower limb ischemia.The pseudo-operation group was given only exposure of the femoral artery.Herbs group was treated with the decoction of Chaihu Shugan Powder combined with Dahuang Zhechong Pills for 7 continuous days after the operation.The changes of peripheral blood levels of aspartate aminotransferase（AST）,creatine kinase（CK）,lactate dehydrogenase（LDH）,α-hydroxybutyrate dehydrogenase（HBDH） as well as the double-fluorescence positive cell percentage（cells with CD34 and VEGFR2 positive fluorescence expression） were observed before operation and on the 1st,3rd and 7th day of the operation.Results After operation,peripheral levels of AST,CK,LDH and HBDH increased in the model group as compared with pseudo-operation group,and decreased on the 3rd and 7th day of the operation in the herbs group as compared with those in the model group and with those on the 1st day of operation（P0.05）.The double-fluorescence positive cell percentage was higher in herbs group and model group than that in pseudo-operation group（P0.05）,and the increase was obvious in herbs group（P0.05 or P0.01）.Conclusion Therapy of soothing liver and activating blood can improve the lower limb ischemia in rats,and its mechanism is probably related with the increase of endothelial progenitor cells in peripheral blood vessels.
