摘要
目的观察过表达外源性CCN1对大鼠放创复合伤愈合的影响,并初步探讨其机制。方法利用AdCCN1感染CHO细胞及放创复合伤大鼠创面,Western blot法鉴定CCN1在CHO细胞的分泌和创面组织细胞中的表达,感染后7d取创面组织制作石蜡切片,HE染色观察创面愈合情况,免疫组化检测MMP1、TIMP1、bFGF等创伤愈合相关因子表达。结果 CCN1在CHO细胞和创面组织中均高表达,与AdRFP组相比,AdCCN1组创面成纤维细胞增多,微血管密度增高,MMP1、TIMP1、bFGF等创伤愈合相关因子表达均升高。结论 CCN1具有促进放创复合伤愈合的作用,其机制可能与CCN1促进成纤维细胞增殖和迁移,促进血管形成,并且调控创伤修复相关因子表达有关。
Objective To observe the effects of over-expressed exogenous CCN1 on healing process in rats with combined radiation and wound injury and to explore the initial mechanism.Methods CHO cells and raw surface of rats with combined radiation and wound injury were infected with AdCCN1.Then the expression of CCN1 in CHO cells and wound tissue was measured by Western blot.Seven days after infection,the wound tissue of rats was obtained to make paraffin section.The healing condition of wound tissue was observed after HE staining,and the expression of MMP1,TIMP1 and bFGF was detected by immunohistochemistry.Results CCN1 was found highly expresseion in CHO cells and wound tissue.Compared with AdRFP control group,the fibroblast number and microvessel density of wound tissue increased in AdCCN1 group,and the expression levels of MMP1,TIMP1 and bFGF also increased in the same group.Conclusion CCN1 can promote the healing process of combined radiation and wound injury,and the mechanism may be involved in its promotion of vasiformation,fibroblast proliferation and migration as well as its regulation of the expression of wound repair-related factors.
