“脾制水”与肾小管上皮细胞线粒体的相关性研究

Study of the Relation of "Spleen Restrains Water" and Mitochondriain Renal Tubular Epithelial Cells-Series Study of "Hypothesis of the Essence of Five Zang in TCM"

目的:验证的"中医藏象脾的实质是线粒体"假说中关于"脾制水"的理论的正确性和前瞻性。方法:采用5/6肾切除大鼠CRF模型,随机分为模型组、大黄治疗组、培土制水方大、小剂量治疗组,观察各组Scr、BUN、MDA、SOD和肾脏超微结构(尤其是线粒体)的改变。结果:培土制水方大、小剂量组的Scr、BUN和血浆自由基的水平和肾脏超微结构、肾小管上皮细胞线粒体的损伤均显著轻于模型组和大黄组,(P0.01或P<0.05);大鼠血浆自由基的水平与Scr、BUN呈非常显著正相关,并与肾脏超微结构、肾小管上皮细胞线粒体的损伤程度相一致。结论:培土制水方通过"培土",保护了肾小管上皮细胞的线粒体,所以明显减轻了肾的病理损伤程度和改善了肾功能,达到了"制水"的目的。这也初步验证了"中医藏象实质细胞生物学假说"中关于"脾制水"的微观实质是肾小管上皮细胞线粒体的推论。

Objectives：To verify the correctness and perspectiveness of＂Spleen RestrainsWater＂ in the theory of＂the Mitochondrion is the Essence of the Spleen In Five Zang In TCM＂ that we have put forward.Methods：40 Sprague-Dawley Rats （SD Rats） were divided into five groups at random.8 rats served as normal control group.The remaining rats were given 5/6 subtotal nephrectomies to establish the model of CRF.One month after nephrectomy the model of CRF rats were subdivided randomly into 4 groups：the model control group,the Chinese Rhubarb group,the large,small dose of Soluble Granules Peituzhishui.The animals of large dose received oral administration of Soluble Granules Peituzhishui in a dose of 20.0 kg·d^-1for two months,the small in a dose of 10.0 kg·d^-1,and the Chinese Rhubarb were given Granules of Chinese Rhubarb a dose of 0.21 kg·d^-1（equal to its raw material of a dose of 0.5 kg·d^-1） in the same time.Results：（1） Large and small dose group of Soluble Granules Peituzhishu were significantly lighter than the model group and rhubarb group,with Scr,BUN and plasma levels of free radicals and the degree of renal ultrastructure damage,especially mitochondrial of renal tubular epithelial cell （P〈0.01 or P〈0.05）.（2） The analysis of correlation showed that the BUN and Scr level was positively correlated to that of free radicals.It is consistent with the variations of renal ultrastructure,especially mitochondria of renal tubular epithelial cell.Conclusion：With the therapy principle of ＂strengthen Earth＂,the Soluble Granules Peituzhishu protects and improve the function of mitochondria of renal tubular epithelial cell.In this way,it obviously reduced the degree of renal pathological injury and improved renal function—＂Restraining Water＂.It verify the correctnes of inference from ＂Hypothesis of the Essence of Five Zang in TCM＂ that the substance of ＂Spleen Restrains Water＂ is the renal tubular epithelial cell mitochondria.：
