NF-κB通路对MPTP慢性帕金森病模型早期多巴胺神经元Bcl-xL表达的双重调节

摘要 目的研究NF-κB通路依赖的Bcl-xL表达在1-甲基4-苯基-1,2,3,6-四氢吡啶(MPTP)慢性帕金森病(PD)模型早期DA神经元中的表达过程,探讨NF-κB通路在PD发病机制中的可能作用。方法选择雄性C57BL/6小鼠,随机分为对照组和模型组及干预组,而每组于末次MPTP注射时间点算起,分为12、24h、1及2w4个亚组(n=5)。动态观察小鼠的行为学和黑质Bcl-xL表达及TH阳性神经元数目的变化及NF-κB抑制剂PDTC对上述结果的影响。结果NF-κB通路的效应呈时间依赖性,模型组在12~24h时,Bcl-xL蛋白和mRNA表达较对照组增加(P<0.05),在1~2w时,Bcl-xL表达较前者及对照组减少(P<0.05);在2w时,模型组TH阳性细胞数较对照组减少(P<0.05),而干预组阳性细胞较模型组增多(P<0.05)。结论NF-κB通路对慢性PD模型早期多巴胺神经元Bcl-xL表达具有双重调节作用。
