摘要
目的观察神经生长因子在种植体周围骨愈合中的表达,探讨神经生长因子在种植体周围骨愈合中的作用。方法选用17只SD大鼠,其中16只按观察时间不同随机分为四组。每组大鼠左侧后大腿暴露股骨植入种植体设为实验组,右侧后大腿暴露至股骨设为对照组。术后取股骨标本拍X线片,脱钙后去种植体制作石蜡切片行HE染色、神经生长因子免疫组化染色;剩余1只SD大鼠取颌下腺作为阳性对照。结果种植体植入后均无松动,X线片示各时间组种植体-骨结合良好。HE染色示实验组种植体-骨结合良好,骨愈合过程中可见成纤维组织逐渐减少,1周时可见类骨质,2、3、4周时出现骨小梁,骨小梁逐渐增粗,密度增高。对照组仅骨膜基质细胞和骨髓基质细胞中有神经生长因子阳性染色;实验组1、2、3周时在种植体周围可见较多骨髓基质细胞、骨母细胞、成纤维细胞及成骨细胞神经生长因子阳性染色。4周时较少骨髓基质细胞、骨母细胞阳性染色,未见成纤维细胞有神经生长因子阳性染色。结论NGF表达与骨愈合相关细胞的功能成正比,NGF可能通过调节骨愈合相关细胞的功能来调节种植体周围早期骨愈合。
Obiective To observe the expression of nerve growth factor in bone healing around the implant.Methods Seventeen SD rats were included and divided into four groups (n = 4).One rat salivary gland served as positive control.Implants were implanted in the left thigh,and the right thigh was incised as control.The rats were executed one-four weeks after operation.Specimens were taken to X-ray and decalcified for HE staining and NGF immunohistochemical staining.Results All implants were stable.X-ray showed a good implant-bone ossointegration.NGF staining was positive in periosteum stromal cells and bone marrow stromal cells in the control.NGF positive staining were found in bone marrow stromal cells,periosteal osteoprogenitor,fibroblasts and osteoblasts more in one,two,three weeks and less in four week.The positive staining cells reached the highest rate at two and three weeks after operation.Conclusion Expression of NGF was positively correlated to the activities of bone healing-related cells around the implant.NGF may promote early bone healing around the implant by regulating bone healing-related cells′function.
