环孢霉素A诱导胚胎干细胞向心肌细胞分化

摘要 目的:观察环孢霉素A(CSA)对小鼠胚胎干细胞(ESCs)向心肌细胞分化的影响。方法:分别观察ESCs自发和CSA诱导情况下心肌细胞分化率,细胞免疫组化检测心肌细胞标志物的表达。结果:CSA诱导的心肌细胞分化率明显高于自发的心肌细胞分化率,且诱导生成的心肌细胞具备心肌细胞特有的特征。结论:CSA能够诱导ESCs向心肌细胞分化并扩增ESCs分化形成的心肌细胞。 Objective:To observe the role of cyclosporin-A(CSA)in differentiation of cardiocytes from embryonic stem cells(ESCs).Methods:The differentiation rates of cardiocytes from ESCs with or without CSA induction were observed.The ex-pression of cardiocyte markers were evaluated with immunohistochemistry.Results:Compared with those of spontaneous differentiation,the differentiation rates of cardiocytes were significantly higher in the presence of CSA.CSA-induced car-diocytes showed the specific features of cardiocytes.Conclusion:CSA potently induces cardiocytes derived from ESCs.
