摘要
目的检测血管内皮生长因子(vascular endothelial growth factor,VEGF)、转化生长因子β1(transforminggrowth factorβ1,TGF-β1)在重度子痫前期(Sever pre-eclampsia SPE)患者及正常孕妇母-胎界面表达水平,通过实验结果分析比较两种因子在两组孕妇间表达水平的差异,探索导致患有SPE产妇所产新生儿肺发育较差,NRDS发病率升高的原因,以期最终找到更有效的预防及治疗新生儿呼吸窘迫综合征的方法。方法①实验对象:选取2009年11月至2010年1月在北京妇产医院就诊的患有SPE的孕妇及正常孕妇各20人,两组孕妇均为初产妇,孕妇年龄、孕周及体重无统计学差异,患者对实验知情同意。②实验方法及评估:取母亲外周静脉血、新生儿脐带血、胎盘组织标本一块,采用经典酶联免疫吸附试验检测母血、脐带血、胎盘组织匀浆中的VEGF和TGF-β1两种因子的含量。实验结果采用SPSS13.0软件进行两独立样本的t检验。结果 SPE组与对照组母体外周血VEGF浓度差别无统计学意义(P>0.05);两组脐带血VEGF浓度差别有统计学意义(P<0.05),SPE组高于对照组;两组胎盘匀浆VEGF浓度差别有统计学意义(P<0.05),对照组高于SPE组;两组母体外周血、脐带血、胎盘匀浆中TGF-β1浓度差别有统计学意义(P<0.05),SPE组高于对照组。
Objective: Detect the expression of VEGF and TGF - β1 of two group patients at maternal - fetal interface, then comparison and analysis the difference expression levels of the two groups . Through the results, explore the main reason which lead increased incidence of NRDS of the newborn produced by HDCP mothers. Eventually find a more effective prevention and treatment approach of neonatal respiratory distress syndrome. Methods : ①The choice of experimental samples : The experimental samples is divided into SPE group and normal group. Each group have 20 people, all from the Beijing Maternity Hospital in - patients, both primiparae, two groups of maternal age, gestational age and weight was no significant difference. ②Specimen preparation: Mother peripheral blood, neonatal cord blood and placenta tissue. The concentration of the two factors in maternal blood, cord blood and placental tissue homogenate use classical ELISA assay. The results analysis using spss13.0 software independent samples t - test . Results : There was no significant difference in the concentration of VEGF in matemal blood of two groups ( P 〉 0. 05). The level of VEGF in cord blood was significant differences ( P 〈 0.05 ), SPE group higher than control group. The level of VEGF in placental homogenate was significant differences ( P 〈 0.05), control group higher. The level of TGF - β1 in maternal blood, cord blood and placental homogenate were significant differences ( P 〈 0.05), SPE group higher than control group.
