摘要
目的以电穿孔法,将GFP质粒转染SGC7901/ADM细胞,优化转染条件以得到较高的转染率。方法在400μl电转染体系中,以不同的电场强度(150 V、180 V、210 V和240 V),脉冲时间(5 ms、25 ms、50 ms和75 ms),电场强度和脉冲时间(150 V,75 ms;180 V,50 ms;210 V,25 ms;240 V,5ms),分别将GFP质粒电转染SGC7901/ADM细胞,并检测不同条件下细胞的存活率和转染率。结果 180V/50 ms和210V/25 ms的条件下,均能取得较好的电转染效果。
Objective To optimize the program in which higher transfection rate of the GFP plasmid into SGC7901/ADM cells with electroporation method. Methods SGC7901/ADM cells were transfected in 400μL transfection volume by different poration voltage(150V、180V、210V and 240V),impulse times(5ms,25ms,50ms and 75ms),poration voltage and impulse times(150V,75ms;180V,50ms;210V,25ms;240V,5ms),and the survival rates and transfection rates were calculated.Results The optimal program containing poration 210V /25ms and 180V/50ms will get the best electroporation transfection rate of the GFP plasmid into SGC7901/ADM cells.
