摘要
目的证明酸藤子酚(embelin)可以逆转白血病耐药细胞株K562/D对柔红霉素(DNR)的耐药性。方法应用MTT法求出embelin作用于K562/D24h后的IC10以及K562/D对DNR的耐药指数和逆转倍数;应用annexinⅤFITC/PI复染流式细胞仪检测细胞凋亡;应用JC-1染色流式细胞仪检测线粒体膜电位(△Ψm)。结果K562/D细胞对DNR耐药,耐药指数为7.72;小剂量embelin(5μg/ml)可以逆转K562/D细胞对DNR耐药性,逆转倍数为7.46;embelin(5μg/ml)与DNR(0.1μg/ml)联合作用于K562/D细胞6h后△Ψm即出现明显去极化,细胞凋亡率呈时间依赖性升高,与对照组比较有统计学差异。结论embelin可以通过促进细胞凋亡逆转K562/D对DNR的耐药性,并且在凋亡早期△Ψm已经出现明显去极化,说明凋亡不可逆转。
Objective To illustrate embelin could overcome the resistance of leukemia drug-resistant cell line K562/D to daunorubcin(DNR).Methods IC10 of embelin on K562/D after 24 hours,resistance index and reverse fold were observed by MTT assay.Apoptosis rates of cells were observed by annexin Ⅴ FITC/PI double staining and flow cytometry.Mitochondrial menmbrane potential(△Ψm)were observed by JC-1 staining and flow cytometry.Results K562/D is resistant to DNR and resistance index is 7.72.Small dosage embelin(5 μg/ml)could reverse the resistance of K562/D to DNR and reverse fold is 7.46.△Ψm appears obvious depolarization 6 hours after embelin(5 μg/ml)in combination with DNR(0.1 μg/ml)on K562/D.Increase of apoposis rates is time-dependen(twith statistical significance compared to control group).Conclusion Embelin could overcome the resistance of K562/D to DNR.△Ψm depolarization appears ahead of apoptosis showing that apoptosis couldn't be reversed.
