小叶野决明花粉萌发与低温贮藏研究 被引量:2

摘要 用不同方法测定小叶野决明花粉生活力,确定小叶野决明花粉离体萌发的最适培养基、培养时间、培养温度,并在常温(25℃)、4℃和0℃条件下对小叶野决明花粉进行了低温贮藏研究。结果表明,离体萌发法可以有效测定花粉生活力;花粉在10%蔗糖+1%硼酸的培养基上萌发率最高,25℃培养3 h为花粉的最佳萌发条件,随培养温度和培养时间的增加,花粉萌发力降低;贮藏天数增加,小叶野决明花粉生活力下降;贮藏温度越低,花粉生活力下降越缓慢;0℃下贮藏效果最好。 Performance of different techniques to evaluate the germination of pollens collected from Thermopsis,as well as effects of culture media and preservation temperature and time were studied.The results showed that the culture media is the best method to evaluate the germination of Thermopsis pollen;the pollen germination percentage reached the highest after incubated for 3 hour on the media supplemented with 10% sucrose and 1% boric acid under the 25℃;the pollens germinated at a decreasing rate as the culture temperature increased and culture time prolonged;the pollens germinated at a decreasing rate as their storage prolonged,and the lower their storage temperature,the slower their germination rate declined;0℃ is the most favorite preservation temperature.
