摘要
目的:探讨甲状旁腺素相关肽(PTHrP)对大鼠胰岛β细胞瘤株INS-1胰岛素分泌的促进作用。方法:分别应用不同浓度PTHrP干预INS-1细胞,应用MTT法检测细胞生长活力,然后应用Western blot检测细胞Ki67、PDX-1、Bax及Bcl-2的表达,BrdU免疫荧光检测细胞增殖情况。结果:PTHrP具有浓度依赖性的促进INS-1细胞增殖,PTHrP显著上调Ki67、PDX-1及Bcl-2表达,下调Bax表达,PTHrP干预后细胞BrdU表达增高。结论:PTHrP可以促进胰岛β细胞增殖。
Objective To explore the role of parathyroid hormone-related peptide (PTHrP) in the proliferation of insulinoma cell line INS- 1. Methods INS- 1 cells were treated by PTHrP at different concentrations. Cell viability was detected by MTT; expressions of Ki67, PDX-1, Bax, and Bcl-2 were detected by Western blot ; cell proliferation was assessed by BrdU incorperation assay. Results PTHrP increased the proliferation of the cells in a concentration-dependent manner. It markedly upregulated the expressions of Ki67, PDX-1, and Bcl-2 while downregutated the expression of Bax. The expression of BrdU was significantly elevated after the treatment of PTHrP. Conclusions Parathyroid hormone-related peptide can increase the proliferation of insulinoma cell line INS-1.
