摘要
目的:探讨十溴联苯醚(BDE-209)对体外培养新生鼠神经干细胞增殖的影响。方法:取新生1d的SD大鼠海马组织,进行体外培养并分组,空白对照组,DMSO对照组,实验A组的BDE-209浓度为10μg/mL,实验B组的BDE-209浓度为30μg/mL,实验C组的BDE-209浓度为50μg/mL。实验组分别对海马神经干细胞进行体外培养染毒,染毒72h后分别测量各组神经球直径及数目,并于染毒后24、48、72、96、120h记录各组光吸收度A值。结果:从新生鼠海马分离培养的细胞巢蛋白阳性。染毒72h后测量各组神经球直径及数目,发现随着染毒剂量加大,神经球明显变小,数目变少,差异有显著性(P<0.01)。MTT结果显示随着BDE-209暴露浓度及时间增加海马神经干细胞增殖能力下降(P<0.01)。结论:无血清培养基分离培养的新生鼠海马神经干细胞具有增殖能力,BDE-209可以导致神经干细胞增殖能力发生改变,随BDE-209暴露浓度及时间增加神经干细胞增殖能力有不同程度的下降。
Objective To investigate the effects of BDE-209 on the proliferation of neural stem cells (NSCs) of neonatal rats cultured in vitro. Methods Hippocampal tissue of 1-day old neonatal rats was obtained and cultured in vitro, then the isolated NSCs were assigned to blank control group, DMSO control group which received 1%o DMSO medium, experiment A group which received 10 μg/mL of BDE-209, experiment B group which received 30 μg/mL of BDE-209, and experiment C group which received 50 μg/mL of BDE-209. After 72 hours of being exposed to BDE-209, the diameter and the number of neurospheres in each group was measured. And the light absorption (A) in each group after being exposed to BDE-209 for 24, 48, 72, 96, and 129 hours was recorded. Results The nestin was positive in the cells isolated from hippocampal of neonatal rats. The diameter and number of neurospheres were decreased with the concentration exposed to BDE-209 after 72 h (P 〈 0.01 ) increasing. The proliferation of NSCs showed a dose- and time, dependent tendency, and the proliferative ability of NSCs decreased with the concentration and time exposed to BDE-209 increased (P 〈 0.01 ). Conclusions The NSCs which were isolated from hippocampus of neonatal rats and cultured in serum-free medium have the ability of self-renewal and proliferation, and BDE-209 could negatively change the proliferative capacity of NSCs, with dose- and time- dependency.
