摘要
采用50日龄的雄性"军牧一号"仔猪进行冷刺激试验,分别在冷刺激0、0.5、3、6、12、24h与应激恢复0.5、3、6、12、24、48h共12个时间点采取仔猪肝脏、脾脏和股二头肌样品。利用Western blot检测冷刺激(-7±2℃)不同时间仔猪肝脏、脾脏与股二头肌HSP70的表达量,探讨冷刺激对机体3种主要器官内HSP70表达量的影响。结果显示仔猪肝脏、脾脏和股二头肌中HSP70的表达量都以不同程度的升高,而后呈现先下降再升高的变化趋势。结果提示肝脏、脾脏和股二头肌3种组织尤以肝脏HSP70表达量变化明显,提示冷应激反应中代谢最旺盛的器官肝脏中物质代谢可能与HSP70的表达量有关。
"Junmu 1"piglets aged 50 d were subjected to cold stress at the different time to explore the changes of HSP70.The piglets were in cold environment(-7±2 ℃) for 24 h.The piglets were slaughtered and the samples of liver,spleen,muscles were collected at stress for 0,0.5,3,6,12,24 h and 0,0.5,3,6,12,24,48 h after stressed recovery.HSP70 in liver,spleen and muscles were measured by Western blot to explore the effects of cold stress on HSP70.The results showed that the expression of HSP70 in liver,spleen and muscles increased at the different speed,then had the trend of firstly lowering to elevating.It is concluded that HSP70 in the three tissues especially liver changed obviously,and substance metabolism in liver in cold stress had a relation with the expression of HSP70.
