摘要
目的探讨中药四物消风颗粒对变应性接触性皮炎(allergic contact dermatitis,ACD)小鼠免疫器官胸腺、脾脏及其T、B淋巴细胞增殖的影响。方法将60只小鼠完全随机分为6组,分别为正常对照组,ACD模型对照组,氢化可的松阳性对照组及四物消风颗粒高、中、低剂量组,每组10只。观测各组小鼠耳肿胀程度、胸腺指数、脾脏指数以及丝裂原诱导的脾T、B淋巴细胞的增殖反应。结果与模型对照组小鼠耳肿胀程度比较,氢化可的松组,四物消风颗粒低、中、高剂量组小鼠耳肿胀程度差异均有统计学意义(P<0.05)。两两比较显示四物消风颗粒中、高剂量组优于低剂量组(P<0.05),与阳性对照组比较无明显差异(P>0.05)。与模型对照组小鼠胸腺指数、脾脏指数比较,氢化可的松阳性对照组、四物消风颗粒低剂量组、中剂量组、高剂量组均显著减低(P<0.05),四物消风颗粒高、中剂量组优于低剂量组(P<0.05),四物消风颗粒高剂量组与阳性对照组无明显差异(P>0.05)。与模型对照组脾脏T、B淋巴细胞增殖活性比较,氢化可的松阳性对照组、四物消风颗粒低剂量组、中剂量组、高剂量组差异均有统计学意义(P<0.05)。同时两两比较显示四物消风颗粒中、高剂量组优于低剂量组(P<0.05),与阳性对照组比较无明显差异(P>0.05)。结论四物消风颗粒防治小鼠ACD的药理机制可能与其调节T、B淋巴细胞功能作用有关。
Objective To investigate the effects of four ingredients for dispersing pathogenic wind (FIDPW) granules on thymus,spleen and proliferation of their T and B lymphocytes in mice with allergic contact dermatitis (ACD).Methods Sixty mice were randomly divided into normal group,ACD group,hydrocortisone (HC) group and high,medium,and low FIDPW dose groups,10 mice in each group.Auricle swelling,spleen and thymus indexes,mitogen-induced proliferation of spleen T and B lymphocytes were determined.Results The auricle swelling was lower in HC,low,medium and high FIDPW dose groups than in ACD group (P0.05).The effect of high and medium FIDPW dose was better than that of low FIDPW dose on auricle swelling (P0.05) with no difference in HC,high and medium FIDPW dose groups (P0.05).The thymus and spleen indexes were significantly lower in HC,low,medium and high FIDPW dose groups than in ACD group (P0.05).The effect of high and medium FIDPW dose was better than that of low FIDPW dose on thymus and spleen indexes with no difference between high FIDPW dose and HC groups (P0.05).A significant difference was found in mitogen-induced proliferation of spleen T and B lymphocytes in ACD,HC,low,medium and high FIDPW dose groups (P0.05).The effect of medium and high FIDPW dose was better than that of low FIDPW dose on mitogen-induced proliferation of spleen T and B lymphocytes (P0.05) with no difference between low and high FIDPW dose and HC groups (P0.05).Conclusion FIDPW granules can prevent and treat ACD in mice by regulating the function of T and B lymphocytes.
