β_2肾上腺素受体基因Arg16Gly变异与高糖低脂膳食诱导的健康青年女性血脂及载脂蛋白比值的关联分析 被引量:2

Correlation between β_2-adrenergic receptor gene Arg16Gly variation and blood-lipid and apolipoprotein ratio mediated by high carbohydrate/low fat diet in healthy young females

摘要 目的探讨β2肾上腺素受体(β2-adrenoceptor,β2AR)基因(β2AR)Arg16Gly多态性对健康青年血脂及载脂蛋白比值的影响及在高糖低脂(high-carbohydrate/low-fat,HC/LF)膳食诱导的血脂及载脂蛋白比值变化中的作用。方法56例健康志愿者,给予7d平衡膳食和6dHC/LF膳食,于第1、8天及第14天取12h空腹静脉血,测定血脂和载脂蛋白(apolipoprotein,apo)AI、B100水平,计算TG/HDL-C、log(TG/HDL-C)、TC/HDL-C、LDL-C/HDL-C、apoAI/apoB100值。提取基因组DNA,聚合酶链反应-限制性酶切法分析Arg16Gly多态性。结果不同性别AA基因型受试者与G等位基因携带者之间血脂比值的基础值差异无统计学意义(P>0.05)。平衡膳食后及HC/LF膳食后,无论是受试人群整体还是将男女分组分析,AA基因型受试者与G等位基因携带者之间血脂及载脂蛋白比值亦无明显差异(P>0.05)。与HC/LF膳食前相比,HC/LF膳食后,在受试人群整体,AA基因型受试者TG/HDL-C(P=0.017)、log(TG/HDL-C)(P=0.031)、apoAI/apoB100(P=0.006)显著升高,LDL-C/HDL-C和TC/HDL-C显著降低(P<0.01),而G等位基因携带者仅LDL-C/HDL-C和TC/HDL-C显著降低(P<0.01)。男女分组分析发现,与HC/LF膳食前相比,男性受试组中HC/LF膳食后LDL-C/HDL-C(AA基因型:P=0.035,G等位基因:P=0.002)和TC/HDL-C(AA基因型:P<0.01,G等位基因:P<0.01)在AA基因型受试者与G等位基因携带者中均显著下降,apoAI/apoB100仅在AA基因型显著升高(P=0.003);女性受试组中,HC/LF膳食后LDL-C/HDL-C(AA基因型:P=0.011,G等位基因:P=0.043)和TC/HDL-C(AA基因型:P<0.01,G等位基因:P<0.01)在AA基因型受试者与G等位基因携带者均显著下降,TG/HDL-C(P=0.001)和log(TG/HDL-C)(P=0.001)仅在AA基因型较膳食前显著升高。结论β2肾上腺素受体基因Arg16Gly变异能抑制HC/LF膳食诱导的健康青年女性TG/HDL-C和log(TG/HDL-C)升高,而HC/LF膳食诱导的TG/HDL-C和log(TG/HDL-C)改变在该位点野生型(AA基因型)和变异携带者(G等位基因)健康青年男性之间没有明显差异,可能与apoAI/apoB100有关。 Objective To study the effects of β2-adrenergic receptor gene Arg16Gly polymorphism on blood lipid and apolipoprotein ratio and its role in blood lipid and apolipoprotein ratio mediated by high carbohydrate/low fat (HC/LF) diet in healthy young persons.Methods Fifty-six healthy young volunteers had regular diet for 7 d followed by HC/LF diet for 6 d.Twelve-hour fasting venous blood samples were collected on days 1,8 and 14 to measure blood lipid and apolipoprotein (apo) AI and B100 levels,and to calculate ratios of TG/HDL-C,log (TG/HDL-C),TC/HDL-C,LDL-C/HDL-C and apoAI/apoB100.DNA was isolated from genome.Arg16Gly polymorphism was analyzed by PCR-RFLP.Results No significant difference was found in the baseline lipid and apolipoprotein ratio in subjects with AA genotype and G carriers before and after regular or HC/LF diet.The ratios of TG/HDL-C (P=0.017),log (TG/HDL-C) (P=0.031),and apoAI/apoB100 (P=0.006) were significantly higher,while those of TC/HDL-C (P=0.001) and LDL-C/HDL-C (P0.01) were significantly lower in subjects with AA genotype after HC/LF diet than before HC/LF diet.The ratios of TC/HDL-C (P=0.001) and LDL-C/HDL-C (P0.01) were significantly lower in G carriers after HC/LF diet than before HC/LF diet.The ratios of LDL-C/HDL-C (P=0.035) and TC/HDL-C (P0.01) were significantly lower in male subjects with AA genotype and G carriers,while those of TG/HDL-C(P=0.001) and log (TG/HDL-C)(P=0.001) were higher in female subjects with AA genotype and G carriers after HC/LF diet than before HC/LF diet.Conclusion β2AR Arg16Gly gene variation can inhibit HC/LF diet-mediated TG/HDL-C and log (TG/HDL-C) increase in healthy young females after HC/LF diet.There is no significant difference in TG/HDL-C and log (TG/HDL-C) between males and females with AA genotype and G carriers before and after HC/LF diet,which may be associated with the apoAI/apoB100.
