利钠肽C受体在利钠肽舒张猪冠状动脉血管中的作用 被引量:1

摘要 目的:探讨利钠肽C受体是否参与3种利钠肽的舒张血管作用。方法:采用离体血管环灌流方法,观察心房利钠肽(ANP)、脑钠肽(BNP)、C型利钠肽(CNP)、硝酸甘油和利钠肽C受体激动剂cANF4-23对猪冠状动脉的作用;然后加入利钠肽C受体阻断剂cANF4-28观察利钠肽舒张血管作用的变化。实验分为8组(n分别=8):分别为ANP组、BNP组、CNP组、硝酸甘油组、cANF4-23组、cANF4-28+ANP组、cANF4-28+BNP组和cANF4-28+CNP组。结果:CNP组冠状动脉最大舒张率(36.5±4.0)%低于ANP组(60.4±22.6)%、BNP组(68.5±11.5)%和硝酸甘油组(53.8±6.9)%,差异均有统计学意义(P均<0.05);其余各组间比较差异无统计学意义(P均>0.05)。cANF4-28+CNP组最大舒张率为(21.3±10.3)%与CNP组(36.5±4.0)%比较冠状动脉最大舒张率明显减弱(P<0.05),差异有统计学意义。cANF4-23组冠状动脉最大舒张率为(42.4±17.6)%,与CNP组相比差异无统计学意义(P>0.05)。结论:3种利钠肽对猪冠状动脉均有良好的扩张作用;利钠肽C受体参与了CNP对猪冠状动脉的舒张作用,而没有参与ANP和BNP的扩张冠状动脉作用。 Objective:To investigate the possible involvement of C-type natriuretic peptide receptor(NPR-C) on swine coronary artery di- lation mediated by three kinds of natriuretic peptides(NPs). Methods:In vitro pharmacological investigation was conducted in isolated vascular ring from swine coronary artery. The experiment was designed into 8 groups. Atrial natriuretic peptide (ANP group) , brain natriuretic peptide( BNP group), C-type natriuretic peptide ( CNP group ), nitroglycerin ( NTG group ) ; NPR-C agonist cANF4-23 group ; in addition, NPR-C antagonist cANF4-28 was used as cANF4-28 + ANP group, cANF4-28 + BNP group and cANF4-28 + CNP group. The relaxant effects of three NPs and NTG were examined,the effect of NPR-C "agonist cANF4-23 was observed and the effects of NPs with cANF4-28 were evaluated on swine coronary artery respectively. Resuhs :Vasorelaxant activities of ANP, BNP,CNP andNTG were(60. 4 ±22. 6)%, (68. 5 ± 11.5)%, (36. 5 ±4. 0)% ,and (53.8 ± 6. 9)%, respectively, relaxant response of swine coronary artery to CNP was lower than the others, (P 〈 0. 01 respectively). NPR-C antagonist could inhibit the vasorelaxant activity of CNP. NPR-C agonist could produce the similar relaxant activity of CNP. Conclusion : Three kinds of NPs presented significant vasodilatory effects on swine coronary artery. NPR-C was involved in the effect of CNP dilation of swine coronary artery.
