摘要
目的探讨端粒酶(the human telomerase gene,hTERC)基因在不同宫颈脱落细胞中的表达及与人乳头瘤病毒(humanpa pilloma virus,HPV)感染的关系。方法 2007年10月至2008年10月在中国医科大学附属第四医院采用荧光原位杂交(fluorescent in situ hybridization,FISH)方法 ,检测120例子宫颈脱落细胞(正常宫颈20例、宫颈上皮内瘤变(CIN)128例、CIN227例、CIN317例及浸润性宫颈癌28例)hTERC的表达情况及并进行HPV-DNA分型测定。结果 hTERC基因在正常宫颈、CIN1、CIN2、CIN3及浸润性宫颈癌中的阳性表达率分别为10.00%、21.42%、55.60%、88.24%及92.86%。hTERC基因表达宫颈癌组与CIN各组及正常对照组比较均有统计学意义(P均<0.05),CIN2、3组与正常对照组比较有统计学意义(P<0.01)。宫颈病变与HPV感染的宫颈上皮细胞中CIN1、CIN2、CIN3及宫颈癌的hTERC基因扩增阳性率分别为33.33%、66.67%、87.50%、92.86%,77例HPV阳性患者中,hTERC基因扩增比例为76.62%(59/77);而在23例HPV阴性患者中,hTERC基因扩增比例仅为17.39%(4/23),P<0.05,差异具有统计学意义。结论 hTERC基因的扩增与宫颈病变程度及HPV感染呈正相关,hTERC基因高表达在宫颈癌发生发展中可能起重要作用。
Objective To study the relationships between Human papilloma virus and the human telomerase gene of the women with cervical lesions. Methods Fluorescence in situ hybridization (FISH) were used to detect the expression of hTERC gene in 120 cases of exfoliated cells of cervix (20 cases of normal cervix, CIN1 28 cases, CIN2 27 cases, CIN3 17 cases and 28 cases of invasive cervical cancer). And detect the HPV-DNA of cervix tissue by using flow-through hybridization and gene chip technology in the Forth Affiliated Hospital of China Medical University from Oct. 2007 to Oct. 2008. Results hTERC gene in normal cervix, CIN1, CIN2, CIN3 and invasive cervical carcinoma, the positive expression rates were 10. 00%, 21.42%, 55.60%, 88.24% and 92. 86%. hTERC gene expression in cervical cancer group and CIN groups and normal control group were significantly different (P 〈 0. 05 ) , CIN2 and CIN3 group compared with the normal control group, there was significant difference (P 〈 0.01 ). In cervical lesions and cervical epithelium which infected with HPV, CIN1, CIN2, CIN3 and cervical cancer, amplification of hTERC positive rates were 33.33%, 66.67%, 87.50%, 92. 86% ,comparison between groups,P 〈0. 05, the difference of positive rate about hTERC gene expression of each group has a statistically significant. 77 cases of HPV-positive patients, hTERC gene amplification in 59 cases, the ratio was 76. 62% while in 23 cases of HPV-negative patients, hTERC gene amplification in 4 cases, the ratio was only 17. 39% (P 〈0. 05), HPV infection group and non-infected group comparison, the difference has a statistically significant). Conclusion hTERC gene amplification, the degree of cervical lesions and HPV infection are positively correlated. High expression of hTERC gene may play an important role in the development of cervical carcinoma.
