摘要
目的:探讨肝纤维化组织学量化诊断方法。方法:用二甲基亚硝胺(DMN)诱导SD大鼠肝纤维化模型,常规肝组织切片、Masson三色和苦味酸-天狼红染色,苦味酸固绿-天狼红染色比色测定,图像定量处理和半定量计分分析及羟脯氨酸(Hyp)含量测定,结果:切片胶原比色测定、图像定量处理和半定量计分分析均与肝纤维化程度变化相一致,与肝组织Hyp含量具有良好的相关性,r分别为0.94、0.89和0.88,P均<0.01。结论:切片胶原测定、图像定量处理和半定量计分可作为确定肝纤维化程度的量化指标。
Aim: To discuss the histological quantitative diagnosis methods of hepatic fibrosis. Methods: Using dimethylnitrosamine (DMN) to produce a SD ral: model for hepatic fibrosis, the histological technique was be used and liver hydroxyproline (Hyp) contents wear be detected. The histology technique included HE stain, Masson stain. Picric acid-Sirus red stain, and Picric acid-Sirus red stain colorimetric technique. Then used picture quantitative analysis technique and semi-quantitative scoring method to analysis the effect of colorimetric determination. Results: There were better correlation between the change of hepatic fibrosis and the colorimetric determination of collagen content. picture quantitative determination, semi-quantitative scoring method. There was better correlatin between Hyp content and determining effects of these three methods. The coefficient 'r' of three effects was 0.94. 0. 89 and 0. 88. respectively (P<0. 01). Conclusion: These results suggsted that it is possible to ust collagen content determination in slice. picture quantitative determination and semi-quantitative scoring for the quantitative diagnosis indices in hepatic fibrosis.
