摘要 目的:开发一种快速精子浓度检测方法,并评价其可行性。方法:根据精子可被亚甲兰溶液染成蓝色,其颜色深浅与精子数量呈正相关原理,以经显微镜计数确认浓度的精子(2000万个/ml)为标准样本,加入亚甲蓝溶液染色,过滤,根据结果颜色,应用现代印刷技术制备出标准色标。以颜色深浅检验标本中精子浓度,对方法中涉及的过滤膜、染色液、染色时间、标准色标等关键参数进行确认,评价试剂盒检测结果。结果:通过对3000份成年男性精液样品检测结果表明,该试剂盒方法的灵敏度、特异性和准确性均在95%以上。结论:本方法所制试剂盒能够实现快速自检,适合医院检查前初步诊断,时间短,操作简单。 Objective:To develop a rapid self-testing method for measuring sperm concentration and determine the feasibility of this method.Methods:The sperm cells could be dyed by methylene blue solution,and the color gradation associated with the sperm number.The sperm samples with the concentration of 20 million /ml set as standard samples were dyed with a prepared methylene blue solution,and filtered to remove the unbound dye.The average code developed from the color gradation of the standard samples served as a standardized color code.The key parameters of this method including the filtering membrane,the staining solution,the duration of staining,and the standardized color code were explored.Results:Verified with 3 000 samples donated by adult male volunteers,the sensitivity,specificity and accuracy of this method were found to be more than 95%.Conclusion:This method with short testing duration and easy operation can be used for rapidly self-test and appropriate for primary diagnosis at home.
