新型血管内栓塞材料原位植烷三醇液晶的生物相容性考察及体内评价 被引量:5

Investigation on Biocompatibility and in Vivo Characteristics of in Situ Phytantriol Liquid Crystalline

摘要 目的制备及评价一种用于血管内栓塞治疗肝癌的新型栓塞材料原位植烷三醇液晶。方法通过三元图实验设计筛选原位植烷三醇液晶的最佳处方,测定其黏度及胶凝时间,并考察此原位植烷三醇液晶的生物相容性;采用数字减影血管造影术检查评价原位植烷三醇液晶的栓塞效果,术后1周取栓塞组织石蜡切片,常规HE染色,光镜观察。结果当水的质量分数小于50%时,所得各向同性溶液具有良好的流动性,并能在体内迅速形成植烷三醇立方液晶;另外,植烷三醇液晶无细胞毒性,不引起溶血,对出血和凝血时间无影响;栓塞实验表明该植烷三醇液晶能有效地闭塞血管,阻断肝动脉的血流,病理检查发现栓塞7 d后植烷三醇液晶仍能滞留于血管中。结论植烷三醇液晶具有良好的生物相容性及栓塞效果,是一种理想的新型栓塞材料。 OBJECTIVE To prepare in situ phytantriol liquid crystalline and evaluate its biocompatibility and embolization effects for the treatment of hepatocellular carcinoma.METHODS Triangular phase diagram was generated to screen the optimum formulation of insitu phytantriol liquid crystalline.The viscosity and sol-gel time were measured.The biocompatibility of phytantriol liquid crystalline was investigated.The hepatic angiographies were performed in New Zealand white rabbits under digital subtracted angiography(DSA) by injecting in situ phytantriol liquid crystalline to observe and evaluate its effects on embolization of rabbit hepatic artery.The sections of embolized tissue were examined microscopically.RESULTS The isotropic solution was flowable,and was transformed into cubic phase rapidly when the percent of water in phytantriol-ethanol-water system was less than 50%.Safety study results demonstrated that phytantriol liquid crystalline was toxicity-free,caused no hemolysis,and showed no influence on bleeding time and clotting time.In vivo,in situ phytantriol liquid crystalline effectively blocked the vessels and the blood flow.The pathological analysis revealed that the in situ phytantriol liquid crystalline was still in arteries 1 week after embolization.CONCLUSION In situ phytantriol liquid crystalline showed acceptable biocompatibility and excellent embolic effect,which could be a promising liquid embolic agent.
