冠状动脉粥样硬化患者钙结合蛋白S100A11基因表达的变化 被引量:1

摘要 目的探讨外周血单个核细胞钙结合蛋白S100A11基因表达水平与冠状动脉粥样硬化的关系。方法连续入选接受冠状动脉造影的患者114例,结合临床症状和冠状动脉造影结果分为稳定性心绞痛组(61例)和对照组(53例)。冠状动脉造影前抽取患者空腹静脉血进行血常规和高敏C反应蛋白(hs-CRP)检测、提取总RNA并合成cDNA用于实时定量RT-PCR。结果稳定性心绞痛组糖尿病、闭塞性动脉粥样硬化和(或)缺血性脑卒中病史及hs-CRP水平明显高于对照组(P<0.05,P<0.01);稳定性心绞痛组淋巴细胞百分比明显低于对照组(P<0.05),S100A11基因表达水平明显高于对照组(P<0.05)。S100A11基因表达水平与hs-CRP水平呈正相关(R^2=0.05,P<0.05)。结论稳定性心绞痛患者的外周血单个核细胞S100A11基因表达水平升高,提示其持续高表达可能与冠状动脉粥样硬化有关。 Objective To evaluate the expression level of S100A11 gene in peripheral blood mono- nuclear cells(PBMC). Methods Patients were divided into stable angina pectoris(SAP, n = 61) and control (n = 53) groups as documented by coronary angiography(CAG). Venous blood was obtained before CAG for RNA isolation and routine blood count. Expression levels of the genes in individuals were evaluated by quantitative real time RT-PCR. Results High-sensitivity C-reactive protein(hs-CRP) level and rate of atherosclerosis obliterans and/or isehemic stroke history in SAP group were higher than those in the control group (P ~ 0.05,P ~ 0. 01, respectively). S100All expression level was higher in SAP than in the control(1.07±0.05 vs O. 89±0.06, P 0.05) ,but lymphocyte count was lower in the former than in the later(27.2% vs 31.7%, P 0.05). S100A11 expression level was positively correlated with hs-CRP level (R2 = 0.05,P 0.05). Conclusion Up-regulation of S100A11 gene expression in PBMC may be associated with coronary artery atherosclerosis.
