摘要
目的 探讨房颤患者β-纤维蛋白原-455G/A(β-Fg-455G/A)基因多态性与血浆纤维蛋白原(Fg)水平、缺血性脑血管病的关系.方法 应用聚合酶链反应一限制性片段长度多态性方法(PCR-RFLP)检测92例房颤合并脑梗塞患者、80例单纯房颤患者和98名健康者的β-Fg-455G/A基因,并测定血浆Fg水平.结果 ①房颤合并脑梗塞组β-Fg-455G/A的A等位基因频率为0.304,房颤组为0.344,健康组为0.179,房颤合并脑梗塞组和房颤组与健康对照组比较差异有统计学意义(P〈0.05),房颤合并脑梗塞组与房颤组比较差异无统计学意义(P〉0.05).②房颤合并脑梗塞组血浆Fg水平为(3.83±0.59)g/L,房颤组血浆Fg水平为(3.58±0.45)g/L,健康组对照组血浆Fg为(2.79±0.36)g/L,房颤合并脑梗塞组与房颤组、健康对照组比较差异有统计学意义(P〈0.05),房颤组与健康对照组比较差异也有统计学意义(P〈0.05).③房颤合并脑梗塞组、房颤组和健康组A等位基因携带者的血浆Fg水平均明显高于同组G/G基因型者,差别有统计学意义(P〈0.05).结论 β-Fg-455G/A基因的A等位基因与房颤合并脑梗塞及房颤关系密切,并且与血浆高水平的Fg有关,血浆Fg水平与房颤合并脑梗塞密切相关.
Objective To investgate the relationships amonge β-fibrinogen-4β-fibrinogen55G/A genes polymorphism with serum fibrogen level and ischemic cardiovascular and cerebral vascular disease. Motheds Polymerase chain reaction and technique of restriction fragment length porlymorphism (PCR-RFLP) were used to determine the genotypes of β-Fg-455G/A in 92 cases of atrial fibrillation accompanied cerebral infarction, 80 cases of atrial fibrillation alone, and 98 health cases as control group. The alleles of them were also determined. Their serum Fg levels were also measured. Results (1)The frequence of A allele of β-Fg-455G/A in the group of atrial fibrillation accompanied cerebral infarction was 0.304, 0.344 in the atrial fibrillation group and 0.179 in the control group. There were significant difference between that in artrial fibrillation accompanied cerebral infarction group and in health group, also significant difference between that in atrial fibrillation group and in health group (P〈0.05). There was no significant difference between the two diseased groups(P〉0.05 ). (2)The serum Fg level in atrial fibrillation accompanied cerebral infarction group was (3.83±0.59)g/L, (3.58±0.45)g/L in atrial fibrillation group and(2.79±0.36)g/L in health group. The serum Fg level in atrial fibrillation accompanied cerebral infarction group has significant difference compared with that in the atrial fibrillation group and in health group; also signifi- cant difference between that in atrial fibrillation group and health group (P〈0.05), and significant difference be- tween that in atrial fibrillation group and health group (P〈0.05). (3)The serum level of Fg in A allele genotypes was significant higher than that in G/G genotypes in every group (P〈0.05). Conclusion The A allele in gene of 15-Fg-455G/A have close relationship with atrial fibrillation and atrial fibrillation accompanied cerebral infarction, also have close relationship with high serum level of Fg .The serun Fg level has close relationship with the onset of atrial fibrillation accompanied cerebral infarction.
