摘要
目的研究武汉市生活饮用水中有机提取物对人肝肿瘤HepG2细胞DNA损伤作用。方法用XAD-2/8复合树脂(XAD-2和XAD-8树脂等体积混合)等比富集武汉市生活饮用水中有机污染物,应用慧星试验检测有机提取物的遗传毒性。结果该市不同水源、不同水文期下饮用水中有机提取物对HepG2细胞均产生不同程度的DNA损伤作用。同一水文期不同水源比较,在剂量10和30ml/ml下,汉江水源末梢水有机提取物致DNA损伤程度大于长江水源(P<0.05);同一水源不同水文期时比较,在剂量10和30ml/ml下,枯水期有机提取物致DNA损伤程度大于丰水期(P<0.05)。结论武汉市主要水源汉江和长江的氯化饮用水有机提取物对HepG2细胞DNA均有损伤作用,损伤作用的严重程度从大到小为汉江水>长江水,枯水期>丰水期。
[Objective]To study the DNA damage in HepG2 cell exposed to organic compounds extraction of drinking water in Wuhan City.[Methods]XAD-2/8 composite resins(XAD-2 and XAD-8 equal proportion mixed resins)were used to concentrate with equal proportion organic compounds from tap water of Wuhan Han.The genotoxicity of organic compounds was tested by comet assay.[Results]The organic compounds extracted from drinking water at different hydrological period and different water sources could cause varying degrees DNA damage of HepG2 cells.In the same hydrology time,the genotoxictiy of organic compounds at dose of 10 and 30ml/ml in Hanjiang River was stronger than that in Yangtze River(P0.05);while at the same water source,the genotoxicity of organic compounds at dose of 10 and 30ml/ml in low water season was stronger than that at ample flow season(P0.05).[Conclusion]Organic compounds extracts of drinking water from water sources of Yangtze river and Hanjiang river in Wuhan City can induce DNA damage in HepG2 cells.DNA damage levels by organic extracts of drinking water from Hanjiang River were the higher than that from Yangtze River.DNA damage levels induced by organic extracts of drinking water sampled in low water season are higher than that in ample flow season.
