摘要
目的:研制一种检测尿胰蛋白酶原-2(Urinary Trypsinogen-2,TSG-Ⅱ)的新型电流型免疫生物传感器。方法:将Nafion吸附到玻碳电极表面,并通过静电吸附和共价键合作用将硫堇和纳米金进行层层自组装,然后通过形成的纳米金单层吸附TSG-Ⅱ抗体,最后用辣根过氧化物酶封闭电极上的非特异吸附位点。结果:该免疫生物传感器对TSG-Ⅱ的响应特性良好,其线性检测范围为6.0~100μg/L;检出限为3.5μg/L。结论:该电流型免疫生物传感器具有灵敏度高、特异性好等优点,为建立一种检测TSG-Ⅱ新方法奠定了基础。
Objective:To develop a novel amperometic immunosensor for the determination of Urinary Trypsinogen-2(TSG-Ⅱ).Methods:the immunosensor was developed by modifying Nafion film on the glassy carbon electrode(GCE),then thionine and colloidal gold were immobilized by static electricity and covalent bonding self-assembly.And then the antibody of Urinary TSG-Ⅱwas absorbed on the surface of nano-gold monolayer.Finally,Horseradish peroxidase(HRP) blocked the non-specific adsorption site on the electrode.Result:Experiment results showed that the good linear relations between peak current and concentration of TSG-Ⅱ were established in the range of 6.0-100μg/L.The detection limit was 3.5μg/L.Conclusion:The amperometie immunoassay biosensor has high sensitivity and good specificity and it has laid the foundation to establish a new method for detection of TSG-Ⅱ.
