摘要
目的:通过观察干预脐血细胞K+/Cl-离子转运体(KCC)对细胞调节性容量减少(RVD)的生物学改变,为探讨富集胎儿有核红细胞(NRBC)方法提供线索。方法:流式细胞仪计数观察尿素干预脐血KCC转运体后不同密度介质下离心富集NRBC的效果。结果:在高渗400mmol/L尿素干预10分钟条件下,红细胞体积缩小最大(P<0.01)。干预脐血后1.065g/ml密度介质离心下层红细胞白膜层(未干预下层)NRBC富集量由12.20%升为69.90%,富集增长差异有统计学意义(u=7.96,P<0.05),单个核上层(未干预上层)NRBC由7.15%降为2.00%,减少量差异有统计学意义(u=5.47,P<0.05)。结论:尿素干预脐血可以增加有核红细胞富集率,较传统富集法的富集量更高。
Objective:To observe the biological changes of cell regulatory volume reduction(RVD)by intervention of K+Cl-(KCC)in transporter in umbilical cord blood cells,in order to provide clues for the enrichment of fetal nucleated red blood cells(NRBC).Methods:The centrifugal effect of NRBC enrichment in different density media were counted by flow cytometry under urea intervention cord blood KCC transporter.Results:The volume of red blood cell decreased maximum under 400 mmolL hypertonic condition and interfering with urea for 10 minutes(P0.01).Under 1.065 gml density centrifugation medium,the NRBC enrichment of underlayer white film were increased from 12.2% to 69.9%(u=7.96,P0.05)after the urea intervention.The NRBC of upper white film was reduced from 7.15% to 2.00%(u=5.47,P0.05).Conclusions:Nucleated red blood cells are enriched by Urea intervention of umbilical cord blood,which is better than traditional method.
