原生质体诱变选育β-葡聚糖酶高产菌株

Screening of high-yield β-Glucanase producing mutants by protoplast UV mutagenesis

摘要 以β-葡聚糖酶产生菌烟曲霉作为出发菌株,混合酶系作用制备原生质体,经过紫外线和LiCl复合诱变处理,通过对再生平板上长出的诱变菌株进行初筛和摇瓶复筛,最终获得一株酶活较高的菌株UV-26,其活力为3.4U/mL,是出发菌株的3.09倍,并且该菌株具有良好的遗传稳定性。 The protoplasts of Aspergillus fumigatus was prepared by dissolving it with mixed-enzyme system.After the protoplasts were induced by ultraviolet ray and LiCl,a high-yield strain UV-26 was obtained by first screening of flat plate and second screening of shake flask.The enzymatic ability of the strain was increased from the initial 1.1 U/mL to 3.4 U/mL.And it can descend stably after six generation.
