摘要
对1株DHA生产菌裂殖弧菌Schizochytrium sp.的原生质体化及再生条件进行了优化研究,并在此基础上利用代谢途径分析原理,结合紫外诱变,筛选获得DHA的高产菌。实验结果表明:以0.2mol/L磷酸盐缓冲液作为渗透压调节剂,0.3g/L蜗牛酶在30℃下作用20min,原生质体化率可达77.78%,再生率为0.57%。在此基础上,进行紫外诱变3min,获得了1株DHA高产菌Schizochytrium sp.HX-204M,其细胞干质量、脂肪酸产量较出发菌株分别提高了12.27%、13.76%,并且DHA占脂肪酸的百分含量提高了8.43%。
This article offered an experimental analysis of the protoplastization and regeneration of Schizochytrium sp.,aiming to get a high yield production strain of DHA.The investigation showed that the ideal result would be got when 0.3 g/L snailase reacted 20 min with cells at 30 ℃,the protoplastization rate could reach 77.78%,and the regeneration rate reached 0.57%.The mutant,called Schizochytrium sp.HX-204M,was obtained by protoplast mutagenesis,combining with metabolic pathway analysis.Its cell dry weight and fatty acid was increased by 12.27% and 13.76%,respectively.In addition,the DHA content was higher than the control.
