摘要
目的探讨抗人T淋巴细胞猪Ig对再生障碍性贫血小鼠骨髓象及骨髓粒系、红系和巨核系的影响。方法 ICR小鼠60只,雄性,3~4周龄;体质量16~18g;随机分为A组(1000mg·kg-1受试品)、B组(500mg·kg-1受试品)、C组(250mg·kg-1受试品)、D组(1000mg·kg-1阳性对照组)、E组(模型组)、F组(9g.L-1盐水组),每组10只。小鼠皮下注射2.5g.L-1苯玉米油溶液复制再生障碍性贫血模型,骨髓涂片Wright染色,光镜大体观察;油镜下进行骨髓涂片分类计数、骨髓细胞混悬液有核细胞计数。结果 1000mg·kg-1受试品组骨髓片有核细胞多数呈增生活跃状态,部分呈增生减低趋势;500mg·kg-1受试品组骨髓片有核细胞增生减低;250mg·kg-1受试品组骨髓片有核细胞增生明显减低,骨髓小粒造血细胞减少;模型组骨髓片有核细胞增生极度减低,骨髓小粒造血细胞明显减少,但500mg·kg-1、250mg·kg-1受试品组和模型组的淋巴细胞数量较9g.L-1盐水组稍增多,纤维细胞等非造血细胞增加;1000mg·kg-1阳性对照组结果与1000mg·kg-1受试品组结果相同。骨髓有核细胞计数显示,与模型组比较,高、中剂量的受试品组及阳性对照组骨髓有核细胞数明显增加。结论抗人T淋巴细胞猪Ig可明显改善再生障碍性贫血动物的骨髓象。
Objective To explore the influence including bone marrow image, granulocytes, on granulopoiesis, erythropoiesis and megakaryopoiesis of mice induced by anti - human T lymphocyte porcine immunoglobulin. Methods The male ICR mice of 3 - 4 weeks with 16 - 18 g of weight were randomly divided into 6 groups:group A( 1 000 mg · kg^-1 testing drug) ,group B(500 mg · kg^-1 testing drug) ,group C ( 250 mg · kg^-1 testing drug), group D ( 1 000 mg · kg^-1 positive control), group E ( model group ), and group F ( 9 g · L^-1 PhyS), 10 mice in each group. The aregenerative anemia model on the oleum maydis 2.5 g · L^-1 were duplicated by hypodermic injection, the bone marrow morphous were observed by Wright staining, differential counted the bone marrow slides and counted the karyocyte of ;bone marrow cell susp ension. Results Most the karyocytes appearanced the active hyperplastic state except the minority karyocytes appearanced the reduced hyper- plastic state in the 1 000 mg · kg^-1 testing drug group; karyocytes appearanced the reduced hyperplastic state in 250 mg · kg^-1 testing drug group, parvule hematopoietic cell reduced in the bone marrow; karyocytes appearanced the exceeding reduced hyperplastic state in model group, parvule hematopoietic cell reduced in the bone marrow; the amount of achroacyte and others unhematopoietic cell such as fibrocyte had a slightly improvement in 500mg · kg^-1 and 250mg · kg^-1 testing drug group than that in the 9 g·L^-1 PhyS group;the same result was found in the 1 000 mg · kg^-1 positive control group and 1 000mg · kg^-1 testing drug group. Compared to model group, high dosage, karyocytes appearanced the improvement state in the model group middle dosage testing drug group and positive control group. Conclusion Anti - human T lymphocyte porcine immunoglobulin can obviously improve the bone marrow image of mice with aregenerative anemia.
