RNAi干扰HPV16E6基因对宫颈癌Siha细胞的影响 被引量:2

摘要 目的:探讨E6 SiRNA对HPV16阳性Siha细胞E6基因的抑制作用,以及对细胞凋亡的影响。方法:合成两对针对HPV16 E6基因的特异性SiRNA及一对阴性对照SiRNA,脂质体介导转染Siha细胞系。RT-PCR检测各组HPV16 E6基因mRNA表达,Westernblot检测HPV16 E6及P53蛋白变化,流式细胞术检测各组细胞的凋亡率。结果:与未转染组相比,两对SiRNA分别降低E6 mRNA表达水平84.7%和82.3%(P<0.001),同时分别降低HPV16E6蛋白表达水平61.3%及62.4%(P<0.001),另外,P53蛋白表达水平分别增加244%及272%(P<0.001),但两转染组间差异无统计学意义(P>0.05)。两转染组细胞凋亡率分别为(34.38±3.14)%和(11.24±1.88)%,两转染组间细胞凋亡率差异亦有统计学意义(P<0.001)。结论:RNAi明显抑制了HPV16E6基因的转录及翻译,能够提高P53表达,促进细胞凋亡。
